摘 要:目的观察百草枯急性中毒后不同时间段引起血浆以及肺、肾组织中炎症因子TNF-α和IL-10的变化,为探讨百草枯中毒引起的脏器损害机制提供理论依据。方法以20%百草枯溶液(30 mg/kg)制作大鼠中毒模型(PQ组),分别观察模型大鼠在造模2 h,6 h,12 h,24 h,72 h以及120 h其血浆以及肺、肾组织中肿瘤坏死因子-α(TNF-α)和白细胞介素-10(IL-10)水平的变化情况。同时用相同剂量生理盐水腹腔注射做为对照组。结果在PQ中毒大鼠血浆、肺脏、肾脏TNF-α水平在72 h内随时间逐渐升高,在120 h下降;大鼠血浆、肺脏、肾脏IL-10水平在120 h内随时间逐渐升高。PQ组血浆、肺脏TNF-α水平在6 h,12 h,24 h,72 h,120 h与对照组同时段相比显著升高(P<0.05);而PQ组肾脏TNF-α水平在12 h,24 h,72 h,120 h显著高于对照组(P<0.05)。PQ组血浆、肺脏IL-10水平在12 h,24 h,72 h,120 h与对照组同时段相比明显升高(P<0.05);而肾脏IL-10水平在24 h,72 h,120 h与对照组同时段相比有显著增加(P<0.05)。结论百草枯中毒可以引起肺、肾组织损伤,其损伤程度与TNF-α和IL-10水平变化关系密切,提示这种损伤的发生可能与TNF-α和IL-10参与机体的炎症反应和细胞免疫密切相关。Objective To observe the changes of levels of TNF-α and IL-10 in plasma,lung and kidney of the acute poisoned rats induced by paraquat for exploring the mechanism of organ damaged by paraquat poisoning. Methods The rats were intraperitoneally injected with 20% paraquat solution(30 mg/kg)in PQ group and injected with saline in control group.The levels of tumor necrosis factor-α(TNF-α)and interleukin-10(IL-10)in plasma,lung and kidney were determined after poisoning rats for 2 h,6 h,12 h,24 h,72 h and 120 h. Results The levels of TNF-α in plasma,lung and kidney increased gradually within 72 h in PQ group,and decreased at 120 h.TNF-α levels in plasma and lung were significantly higher at 6 h,12 h,24 h,72 h,120 h in PQ group than that in control group(P0.05),and the levels of TNF-α in kidney were significantly higher at 12 h,24 h,72 h,120 h in PQ group than that in control group(P0.05).IL-10 levels increased gradually within 120 h in plasma,lung,kidney in rats.At 12 h,24 h,72 h,120 h,the IL-10 levels in plasma,lung of PQ rats were statistically higher than that in control rats(P0.05).The levels of IL-10 in kidney of PQ rats were significantly higher at 24 h,72 h,120 h than that in control rats(P0.05). Conclusion Paraquat poisoning can cause the damages of lung and kidney,and the damage degrees are closely related with the changes of TNF-α and IL-10 levels,which suggests that the damage may be related with the involvement of TNF-α and IL-10 in the inflammatory response and cellular immune.
