摘 要:目的 研究极低频磁场辐照与肿瘤发生的关系及其潜在的作用机制。方法 应用Northern 及dot 印迹法检测人恶性淋巴瘤Daudi 细胞受不同强度的50 Hz 磁场辐照和( 或) 促癌剂———佛波酯(TPA) 的作用后其原癌基因cfos 的转录水平。结果 0 .8 m T 及0 .4 m T50 Hz 磁场和TPA 处理的cfos基因转录水平分别是单独TPA 处理的143 .4 % 及160 .7 % ( P < 0 .05) 。然而,0 .2 m T 及0 .1 m T磁场则未发现有协同TPA 对cfos 基因的诱导作用( P> 0 .05) 。单独该磁场亦无影响cfos 基因转录的作用( P > 0 .05) 。结论 0 .8 m T 及0 .4 m T50 Hz 磁场对TPA 诱导的cfos 基因转录具有协同促进效应。Objective To study the relationship between extremely low frequency(ELF) magnetic field(MF) and the development of human cancer and its mechanism. Methods Using Northern and dot blot,c fos gene transcription was measured in Daudi cells after exposure to 50 Hz MF at four different flux densities for 20 min,in the presence or absence of phorbol ester(TPA). Results TPA could obviously induce c fos expression in Daudi cells(P<0.05);TPA induced expression of c fos was altered by MF exposure at 0.4 and 0.8 mT(P<0.05).No additive or synergistic induction of c fos expression was observed when combined with MF at 0.1 or 0.2 mT(P>0.05);no changes in c fos expression were found when cells were exposed to MF alone at different flux densities(P>0.05). Conclusion 50 Hz MF exposure could synergize TPA induction in c fos gene transcription under certain flux densities.
