辛伐他汀对脂多糖刺激人脐静脉内皮细胞Tribbles表达的影响

摘　　要：目的研究脂多糖(LPS)对人脐静脉内皮细胞(HUVECs)肿瘤坏死因子(TNF-α)和TribblesmRNA表达变化及辛伐他汀对其表达的影响。方法实验分为12 h组和24 h组,各组又分为空白对照组、脂多糖(100 ng/ml)组、辛伐他汀(5μg/ml)组、辛伐他汀(5μg/ml)和脂多糖(100 ng/ml)共作用组,用实时荧光定量PCR测定LPS刺激后12 h和24 h HUVECsTNF-α和Trb1、Trb2及Trb3的mRNA表达变化。结果与对照组相比,脂多糖组TNF-α、Trb1及Trb3的mRNA表达均明显增加(P均<0.05);与脂多糖组相比,辛伐他汀和脂多糖共作用组TNF-α、Trb1及Trb3的mRNA表达均明显降低(P均<0.05)。结论辛伐他汀可抑制HU-VECs的TNF-α、Trb1及Trb3的表达。Objective To explore the expression of TNF-α and Tribbles mRNA in HUVECs with LPS treatment and the effect of simvastatin on them.Methods HUVECs were cultured and divided into the following experimental groups： control group,LPS group（100ng/mL）,simvastatin group（5μg/mL）,and LPS（100ng/mL）＋ simvastatin（5μg/mL） group for 12h and 24h respectively.The expression of TNF-α,Trb1,Trb2,Trb3 mRNA in HUVECs with LPS treatment after 12h and 24h was measured using real-time quantitative PCR.Results Compared with the control groups,the expression of TNF-α,Trb1 and Trb3 mRNA was significantly increased in LPS groups（P0.05）.However,the expression of TNF-α,Trb1and Trb3 mRNA was statistically decreased in LPS and simvastatin groups compared with LPS groups,（P0.05）.Conclusion Simvastatin could inhibit the expression of TNF-α,Trb1,Trb3 in HUVECs induced by LPS.
