摘  要:建立了同时测定谷子幼苗中赤霉素 A1( G A1)和赤霉素 A3( G A3)的高效液相色谱( H P L C)方法。采用 O D S C18色谱柱,以磷酸盐缓冲液和甲醇为流动相, G A1 和 G A3 在 10 m in 内获得良好分离。利用该方法对谷子的两个正常栽培品种及其矮秆突变体的幼苗进行了测定。测定结果表明,谷子幼苗中同时含有 G A1 和 G A3 两种活性赤霉素,它们各自的质量比是影响谷子株高的重要因素。Gibberellin A 1 (GA 1) and Gibberellin A 3 (GA 3) in millet seedlings were simultaneously determined by HPLC on a Zorbax ODS C 18 (4.6 mm×250 mm,10 μm) column, using a mobile phase consisting of V (0.02 mol/L phosphate buffer solution, pH 4.0)∶V(methanol)=2.2∶1, a flow rate of 0.8 mL/min and a UV detection at 210 nm.GA 1 and GA 3 were separated completely in 10 min. Correlation coefficients for linear equations were 0.998 6 and 0.997 8, recoveries were 102.54% and 99.59%, respectively. Two normal cultivars and their dwarf mutants were determined by this method. The results showed that both GA 1 and GA 3 were discovered in foxtail millet seedling, and endogenous mass ratio of GA 1 and GA 3 in foxtail millet dwarf mutants were significantly lower than those in normal cultivars, and the concentrations of endogenous GA 1 and GA 3 were important factors for plant height.
