摘  要:目的:观察孤儿核受体神经生长诱导基因B-β(nerve growth factor-induced gene B-β,NGFIB-β)在大鼠脊髓发育过程中蛋白表达的动态变化及与增殖细胞核抗原(proliferating cell nuclear antigen,PCNA)的相关关系,探讨NGFIB-β在神经细胞迁移分化中的表达意义。方法:采用石蜡包埋组织切片,免疫组织化学染色及免疫组织化学双重染色。结果:NGFIB-β阳性表达在脊髓的发育过程中呈现出显著的动态变化,NGFIB-β阳性细胞显示已经分化的形态。NGFIB-β首先在胚胎13天(E13)脊髓组织的背侧出现阳性细胞,E15可清楚观察到阳性细胞呈"扶梯"样迁移方式从背侧向腹侧迁移,随着发育和细胞的成熟,阳性表达明显减少,成熟脊髓组织中未见阳性表达,与PCNA的对比观察表明,两种蛋白分别表达在不同的细胞中,NGFIB-β表达在迁移分化的幼稚神经细胞中,而在有增殖能力的细胞中不表达。结论:NGFIB-β可能在大鼠脊髓神经细胞从幼稚到成熟的迁移分化过程中具有调控作用。Objective: To observe the dynamic changes in protein expression of the nerve growth factor-induced gene B-β(NGFIB-β) as well as the relationship between NGFIB-β and proliferating cell nuclear antigen(PCNA) during spinal cord development.And to investigate the significance of NGFIB-β in the differentiating and migrating neural cells.Methods: Paraffin-embedded sections,immunohistochemistry,double labeling immunohistochemical methods were used.Results: The positive expression of NGFIB-β in the developing rat spinal cord presented obviously dynamic changes and the NGFIB-β positive cell presented differentiated form.NGFIB-β was expressed in small quantities in the cells at the dorsal of myeloid tissue in E13.In E15 it was observed that the positive cells migrated towards the ventral side,creating a staircase-shaped image.Along with the maturing of the cells,the positive expression decreased apparently and there was no positive expression of NGFIB-β in mature myeloid tissue.The comparative observation against PCNA showed that the two proteins were expressed at different sites and in different cells.NGFIB-β was mainly expressed in differentiated and migrated immature cells,but not in proliferating cells.Conclusion:NGFIB-β possible plays a regulatory role in the differentiation,migration and maturation of the rat spinal neural cells.
