摘 要:目的探讨bFGF在糖尿病神经病变(DNP)中的作用,为临床提供糖尿病神经病变的形态学依据。方法链脲佐菌素(STZ)60 mg/kg体重一次性腹腔注射,72小时后测尾静脉血糖,血糖≥16.7 mmol/L视为糖尿病模型。在电子显微镜下观察结状神经节超微结构,用免疫组织化学的方法检测bFGF的表达。结果电子显微镜下发现糖尿病神经病变时细胞胞质内线粒体肿胀、溶解、嵴断裂;高尔基体成熟面肿胀;内质网肿胀,明显扩张。细胞核内内质网明显扩张,尼氏体边集。糖尿病大鼠较正常对照组大鼠结状神经节细胞胞浆bFGF表达增强。结论糖尿病时大鼠结状神经节超微结构发生变化,胞浆bFGF表达增强,可能是机体的一种抗损伤反应,bFGF对糖尿病神经病变时的神经元具有修复作用。Objective To explore the effects of basic fibroblast growth factor(bFGF) in diabetes neuropathy(DNP),and offer morphologic data for DNP.Methods The rats in all groups were injected with streptozotocin intraperitoneal(60 mg/kg).Rats with fasting blood glucose≥16.7mmol/L after 72 hours and maintained for 2 weeks were enrolled in DM.The ultrastructural changes of the nodose ganglion were observed by electron microscope,the expression of bFGF was examined by the immunohistochemical method.Results Ultrastructural study showed that the mitochondria,endoplasmic reticulum and Golgi were swollen.The mitochondria was dissolved and its crest was ruptured.The expression of bFGF in cytoplasm of the nodose ganglion in experimental diabetic rats was increased by compared with control group.Conclusion The ultrastructural changes of the nodose ganglion in experimental diabetic rats indicate that neurons are damaged during diabetes.The increasing expression of bFGF may be the response of resistance to the injury of the nerve cells.bFGF gene could effectively enhance recovery of the nerve cells in DNP.
