类风湿关节炎与巨噬细胞炎性蛋白-1α的相关性研究

摘　　要：目的探讨巨噬细胞炎性蛋白-1α(MIP-1α)在类风湿关节炎(RA)患者外周血、关节液的mRNA表达水平及其在RA发病机制中的作用。方法采用实时荧光定量PCR技术检测19例RA、6例骨关节炎患者的关节液和外周血单个核细胞(SFMC和PBMC)中MIP-1αmRNA表达水平。同时采用ELISA法测定MIP-1α的浓度。并与15例健康对照者比较。结果 MIP-1αmRNA在RA组外周血和关节液中均有表达,差异有统计学意义(P<0.01)。RA组MIP-1α的血清及关节液含量明显高于其他组;关节液中MIP-1α的浓度高于血清(P<0.01)。结论 MIP-1α介导的炎性反应是RA发病机制中重要因素。Objective To observe the mRNA expression levels of macrophage inflammatory protein-1α（MIP-1α）in the peripheral blood mononuclear cell（PBMC） and synovial fluid mononuclear cell（SFMC） in patients with rheumatoid arthritis（RA）,and to explore its role in the pathogenesis of RA.Methods The peripheral blood and synovial fluid from 19 patients with RA,6 patients with osteoarthritis（OA） were collected.The real time fluorescent quantitation polymerase chain reaction was used to detect the expression of MIP-1α mRNA in SFMC and PBMC.Results The mRNA expressions of MIP-1α in PBMC and SFMC from patients with RA were 0.60±0.02 and 0.69±0.05.The concentration of MIP-1α in PBMC in RA group,OA group and control group were（200±56）pg/ml,（116±18）pg/ml and（34±12）pg/ml respectively.The levels of MIP-1α in synovial fluid from patients with RA were the highest among the three groups.Conclusion The inflammatory response mediated by MIP-1αis an important factor in the pathogenesis of RA.
