摘 要:目的:观察受体酪氨酸激酶抑制剂genistein对体外培养兔软骨细胞一氧化氮(NO)表达的影响。方法:兔膝关节软骨细胞传代培养,应用不同浓度genistein干预1、2、3、4天后,通过亚硝酸盐测定检测培养上清液NO表达水平。结果:形态学、甲苯胺蓝染色显示3代以内体外培养软骨细胞保持细胞表型的稳定。在genistein干预后上清液中NO水平下降,下降水平与genistein干预浓度和时间相关。结论:体外培养兔软骨细胞三代以内的细胞具有较好的活性,genistein能有效降低兔软骨细胞NO的表达水平。Objective:To investigate the effects of receptor tyrosine kinase inhibitor genistein on the expression of nitric oxide(NO) in chondrocytes of rabbits in vitro.Methods: The passage culture of rabbit articular cartilage cells was carried out with different concentrations of genistein and the cells cultured were divided into 7 groups:control,LPS,and LPS plus to different concentrations genistein(5,10,15,20 and 25μg/mL).The expression of NO was detected in 1,2,3,4 days after intervention.Results: Morphology observation and toluidine blue staining results showed that a stable phenotype within 3 generations of the cartilage cells cultured.The expression of NO was decreased after the intervention of genistein in a concentration and time-dependent manner.Conclusion: Chondrocytes of rabbits could maintain its viability after 3 generation culture in vitro.Genistein can effectively inhibit the expression of NO.
