摘 要:目的:利用近红外光谱法快速测定注射用益气复脉(冻干)中10种人参皂苷的总量。方法:用HPLC分别测定60批样品中10种人参皂苷的总量,运用偏最小二乘法(PLS)建立50批测定值与NIR光谱之间的校正模型,并对10批样品的含量进行预测。结果:运用一阶导数+多元散射校正的预处理方法,选择波段为4 246.8~4 602.2,5 446.8~6 102.6 cm-1,最佳维数为9时,建立的定量模型准确性好,R2(决定系数)和RMSECV(内部交叉验证均方差)分别为94.2,0.186;对10批样品的预测值与HPLC测定值之间的RMSEP为0.234。结论:近红外光谱法能快速对注射用益气复脉中的10种人参皂苷的含量进行预测,方法简便、精密。Objective: To determine the total content of 10 ginsenosides in Yiqifumai lyophilized injection by near infrared spectroscopy.Method: Sixty samples were collected and determined of the total contents of ten ginsenosides by HPLC.The optimal calibration model was established by the contents of 10 ginsenosides in fifty samples and their NIR spectroscopy using the PLS.And the contents of 10 samples were successfully predicted.Result: When using the pretreatment of the first derivative and MSC in the range of 4 246.8~4 602.2,5 446.8~61 02.6 cm-1,the best dimension was 9,and the quantitative model was accurate.The R2 was 94.2,and the RMSECV was 0.186.The RMSEP of ten samples was 0.234.Conclusion: This method is easy,rapaid and precise,and can be used to determine the content of 10 ginsenosides in Yiqifumai lyophilized injection.
