摘  要:以研究东北三省樟子松球壳孢菌Sphaeropsis sapinea的遗传多样性为目的,自黑龙江、吉林、辽宁省不同地区收集、分离获得樟子松球壳孢菌45个菌株,寄主均为樟子松;收集美国菌株2个,分别分离自脂松、班克松。运用形态学和分子生物学手段比较47个菌株的形态学及18S rDNA ITS序列。结果表明:(1)所有47个菌株在形态上没有明显差别;(2)除美国菌株STB1外,其他46个菌株在培养特性上没有明显差别;(3)东北三省的45个菌株之间的遗传距离均小于0.09,与寄主为脂松的A型菌株(STA1)亲缘关系较近,相似度高于98%,与寄主为班克松的B型菌株(STB1)遗传距离为0.13,亲缘关系较远。(4)同一地域、不同地域来源的菌株其ITS序列相同,而地理来源相同的菌株其ITS序列也存在一定的差异;(5)相同寄主上的菌株其ITS序列存在差异,而不同寄主上的菌株其ITS序列相同。综上得出以下结论:东北三省不同地区樟子松上的45个松球壳孢菌菌株为A型。球壳孢菌菌株之间的遗传差异与菌株的寄主及地域性没有明显的关系。In order to study the genetic polymorphisms of Sphaeropsis sapinea on Mongol Scotch Pine(Pinus sylvestris var.mongolica) in the Northeast of China,45 strains were collected from different areas in Heilongjiang,Jilin and Liaoning Provinces;and 2 strains were from American,one was from Red Pine(Pinus resinosa Ait),the other was from Jack Pine(Pinus banksiana Lamb).Morphological and molecular biological methods were used to compare and analyze the morphological characteristics and 18S rDNA ITS sequences of 47 strains.The results showed that(1) there were no significant differences among the morphological characteristics of the 47 strains;(2) there were no significant differences among the cultural characteristics of 46 strains except America strain STB1;(3) the phylogenetic relationship between all 45 northeast strains and American strain STA1 which belongs to type A was closer,the similarity was higher than 98%.And the genetic distances among all 45 northeast strains were less than 0.09.The genetic distance between all 45 northeast strains and American strain STB1 which belongs to type B was 0.13,so the phylogenetic relationship was distant.(4) the ITS sequence of the strains coming from the same area or different areas may be the same,and that coming from the same area may be different;(5) there were differences among the ITS sequence of the strains coming from the same host species,and there were no difference among the ITS sequence of the strains coming from different host species.So the conclusions were(1) Forty-five northeast strains of Sphaeropsis sapinea belonged to A type;(2) To the strains of Sphaeropsis sapinea,there was no significant relationship between genetic differences and regions and hosts.
