摘 要:对采自中国云南省的日本三角涡虫(Dugesia japonica)13个地理种群的细胞色素C氧化酶Ⅰ亚基基因(COⅠ)的部分片段进行PCR扩增、克隆和测序,以采自河南省的日本三角涡虫Luoyang种群为外群,用Clustal X 1.80、MEGA 4.0、PAUP*4.0、MrBayes 3.1.1等程序对其进行序列分析和系统树构建。结果表明:13种群的COⅠ基因片段A+T的含量差异很小,稳定在67.1%~68.8%之间,明显高于G+C的含量。序列间的成对遗传距离介于0.000~0.199之间,其中Yuxi、Dali2、Lincang1 3个种群间的遗传距离最小,为0;Puer和Xishuang种群间的遗传距离最大,为0.199。最大简约树和贝叶斯树拓扑结构基本一致,均以较高支持率聚为两支。结合云南省的地形地貌及丰富的水系特征,推测云南省日本三角涡虫可能是中部起源,扩散至边缘,也可能相反。究竟哪种推测更为可靠,尚待进一步研究。The fragments of COⅠgene of 13 populations of Dugesia japonica collected from Yunnan Province,China were amplified via PCR,and then cloned and sequenced.The sequences were aligned and analyzed with Clustal X 1.80 and MEGA 4.0,and the maximum parisimony(MP) tree and Bayes tree were reconstructed via PAUP*4.0 and MrBayes 3.1.1,respectively.The results show that the A+T contents of 13 sequences have little difference,ranging from 67.1% to 68.8%,more than the G+C contents obviously.The pairwise genetic distances based on model Kimura 2-parameter between sequences range from 0 to 0.199.Distances among populations of Yuxi,Dali2 and Lincang1 were 0,that between Puer and Xishuang was the largest(0.199).The MP tree and the Bayes tree almost show the same topology.Based on the clustering pattern of partial COⅠ genes and the geographical features of Yunnan,it is hypothesized that the freshwater planarian D.japonica in Yunnan Province could have originated from the central sites,then diffused to the marginal sites,or vice versa.More data are needed to test the hypothesis.
