摘 要:目的:探讨纳洛酮对丙泊酚致小鼠学习记忆障碍以及催眠作用的影响。方法:小鼠腹腔注射不同剂量的丙泊酚(10,100 mg·kg-1)建立模型,跳台、避暗实验中观察不同剂量的纳洛酮sc对小鼠跳台潜伏期(stepdown latency,SDL)、步入潜伏期(step through latency,STL)和错误次数的影响;催醒实验中观察不同剂量的纳洛酮皮下注射(subcutaneously,sc)对小鼠睡眠潜伏期和持续时间(sleeping time,ST)的影响。结果:跳台、避暗实验中,用药后第1 d,纳洛酮0.5 mg·kg-1sc能够延长丙泊酚所致记忆障碍小鼠的SDL、STL,减少错误次数(P<0.05;P<0.01),用药后第2、3、4 d,各组SDL、STL和错误次数的差异均无显著性(P>0.05);催醒实验中,纳洛酮0.5 mg·kg-1sc对丙泊酚催眠小鼠的潜伏期和ST没有明显影响(P>0.05),纳洛酮1.2 mg·kg-1sc能够延长丙泊酚催眠小鼠的潜伏期,缩短其ST(P<0.05;P<0.01)。结论:纳洛酮可以改善丙泊酚所致的学习记忆障碍,可以减轻丙泊酚的催眠作用。Objective:To investigate the effect of naloxone on propofol-induced learning deficit and dysmnesia and hypnosis of mice. Methods:Animal models were established by intraperitoneal injecting(ip) of propofol(10 and 100 mg·kg^-1).Normal saline or different doses of naloxone were subcutaneously injected(sc),then the step down latency(SDL),error times in step down test,step through latency(STL),error times in step through test were observed and recorded;and sleeping latency and time(ST) in analeptic test were recorded. Results:In step down test and step through test,the first day after treatment,naloxone 0.5 mg·kg^-1(sc) increased the SDL and STL,and decreased the error times of mice treated with propofol(P0.05;P0.01);2,3,4 days after drug,there was no significant difference in SDL,STL,and error times of each group(P0.05);In analeptic test,naloxone 0.5 mg·kg^-1(sc) on propofol hypnosis in mice did not affect the latency and ST(P 0.05),naloxone 1,2 mg·kg^-1(sc) extend the sleeping latency and decreased the ST(P0.05;P0.01). Conclusions:naloxone could improve propofol-induced learning deficit and dysmnesia and abate the hypnosis of propofol.
