一株鸽新城疫病毒的分离鉴定

摘　　要：对广东地区疑似发生鸽新城疫感染的鸽病料进行病毒分离,通过血凝试验(HA)、中和试验、F基因扩增及序列测定。结果分离到1株血凝效价为4㏒2,且能被NDV阳性血清中和的病毒;用针对NDVF基因设计的特异性鉴定引物对该分离株进行PCR扩增,可扩增出相应的目的片段;测序及Bl ast分析表明其与山东分离株chi cken/Chi na/SD5/2008相似性高达95,%系统进化树分析表明该分离株属于基因Ⅵ型。由此,该分离株为鸽新城疫病毒,并命名为GD2010。A Newcastle disease virus（NDV） was isolated from the pigeon in Guangdong.It was identified NDV with hemagglutination test,neutralization test,PCR and sequence analysis.The HA titer was 4㏒2 and this virus could be neutralized by the standard antibody of NDV.The F gene was amplified by RT-PCR by the specific identification primers of NDV F and sequenced.The homology of F gene was 95% with that of chicken/China/SD5/2008.Phylogenetic analysis showed that this virus was clustered into genotype Ⅵ.So this isolate was identified as NDV and named GD2010.
