摘 要:目的观察受体相关蛋白(RAP)不同区段对被动型Heymann肾炎(PHN)大鼠足细胞瞬时受体电位阳离子通道蛋白6(TRPC6)、synaptopodin、podocalyxin表达和分布的影响。方法给雄性SD大鼠分别注射兔抗RAP全长(RAP—FL组)、氨基端(RAP—N组)和羧基端(RAP—C组)抗血清,建立相应3种PHN模型,对照组注射等量正常兔血清。分别于造模前和造模1周末检测24h尿蛋白量、血清白蛋白和肌酐;造模1周末取大鼠肾皮质行光镜病理及间接免疫荧光激光共聚焦显微镜观察足细胞TRPC6、synaptopodin、podocalyxin的表达和分布。结果造模1周末各模型组24h尿蛋白量显著高于自身造模前和对照组,差异均有统计学意义(均P〈0.01),血清白蛋白和肌酐无明显改变。光镜下各模型组均见肾小球基底膜节段性不均匀增厚,系膜细胞和系膜基质无明显增生。间接免疫荧光激光共聚焦显微镜观察显示,对照组足细胞TRPC6、synaptopodin、podocalyxin沿肾小球基底膜呈均匀、连续线样分布;各模型组TRPC6沿肾小球基底膜呈团块状不均匀分布,荧光强度较对照组有不同程度增强,RAP—FL组较RAP-N组和RAP—C组更为明显,差异有统计学意义(P〈0.05),RAP—N组和RAP—C组间差异无统计学意义(P〉0.05);各模型组synaptopodin和podocalyxin沿肾小球基底膜呈点状、短线状不连续分布,部分节段表达缺失,荧光强度较对照组均有不同程度的减弱,差异均有统计学意义(均P〈0.05);各模型组间差异无统计学意义(P〉0.05)。结论RAP全长及其羧基端与氨基端均存在病理性抗原决定簇,在PHN模型中可上调TRPC6表达,但下调synaptopodin和podocalvxin表达,并改变它们的分布模式,与PHN蛋白尿的发生有关,但在PHN发病中的病理作用仍有待进一步研究。Objective To protein (RAP) on the expression passive Heymann nephritis (PHN). investigate the effects of different sections of receptor associated and distribution of TRPC6, synaptopodin and podocalyxin in Methods Male Sprague-Dawley rats were injected with three kinds of antisera (anti-RAP full-length serum, anti-RAP N-terminal serum and anti-RAP C-terminal serum) to establish three kinds of PHN models. The control group was injected with normal rabbit serum. The quatitation of 24 h urinary protein, serum albumin and ereatinine were taken before injection and one week after PHN model successfully induced. The histopathologic changes of renal tissues were observed by light mieroseopy. The expression and distribution of TRPC6, synaptopodin and podoealyxin in gfomerular podocytes were observed by laser scanning eonfocal microscopy and analyzed by fluorescence quantitative software after indirect immunotluoreseence double staining. Results Tire quantities of 24 h urinary protein in the three model groups were significantly higher than those of themselves before injection and control groups (P〈0.01, respeetively). The values of serum alhumin and ereatinine were not significantly different before and after injection (P〉0.05). The expression of TRPC6 in podocytes was higher in the PHN model groups than that of control group. Fluorescenee intensity of THPC6 in HAP full-length group was stronger than that in HAP N-terminal or C-terminal groups. The expressions of synaptopodin and podoealyxin distributed along the glomerular basement membrane as spot, discontinuous short line and defect of some segments, and were lower in three PHN groups than those of control group. Fluorescence intensity of synaptopodin and podocalyxin among three PHN groups had no differences. Conclusions RAP full-length and N-terminal or C-terminal parts ean inerease the expression of podoeyte TRPC6, but decrease the expressions of synaptopodin and podoealyxin, and alter their distribution, which may be associated with the pmt
