摘  要:目的测定灯盏花乙素和灯盏花乙素苷元表观油水分配系数,研究pH对其影响,为体内吸收研究提供参考.方法采用摇瓶法测定灯盏花乙素和灯盏花乙素苷元的表观油水分配系数,采用HPLC法测定灯盏花乙素和脱灯盏花乙素苷元的浓度.结果 37℃下,pH为3.00,3.50,4.00,5.00,6.00,7.00,8.00,9.00灯盏花乙素的logP分别为-0.10,0.48,-0.37,-0.69,-0.86,-1.33,-0.82,-0.45,-0.28,灯盏花乙素苷元的logP趋近于0.pH对灯盏花乙素的表观油水分配系数有一定的影响,在pH3~7的范围内,随着pH的增加灯盏花乙素表观油水分配系数减小,在pH7~9的范围内又逐渐增大,并且在pH=3.5的磷酸盐缓冲液时表观油水分配系数最大为3.07.结论灯盏花乙素的表现油水分配,受pH影晌较大,而灯盏花乙素苷元的表观油水分配系数受pH影响不大.Objective To provide reference for absorption research by determining the apparent oil-water distribution coefficient of scutellarin and scutrllarein.Method The apparent oil-water distribution coefficient of scutellarin and scutrllarein was determined by shake-flask method and the concentration was determined by high-performance liquid chromatography.Results At 37 ℃,the logP value of scutellarin was-0.10,0.48,0.37,-0.69,-0.86,-1.33,-0.82,-0.45,-0.28 when pH value of the solution was 3.00,3.50,4.00,5.00,6.00,7.00,8.00,9.00,respectively.And the logP value of Scutrllarein was approach 0.Conclusions PH affects the apparent oil-water distribution coefficient of scutellarin.Within pH limits of 3-7,the higher pH,the lower coefficient of scutellarin.And it can reach to the maximum value of 3.07 when the pH of phosphate buffer solution is 3.5,while pH has little impact on the coefficent of scutrllarein.
