摘  要:目的:探讨卵胞浆内单精子注射(ICSI)时注射完整顶体的人精子对卵及胚胎发育的潜在危害。方法:①分别注射1、3、5个顶体完整的人精子于金黄地鼠卵中,同时注射数量相等的无顶体精子作为对照组,观察卵母细胞活化、受精及后期发育情况。②将6 ̄9个顶体完整的人精子注入废弃的人卵母细胞中,观察卵及胚胎发育情况。结果:①将顶体完整的人精子注入金黄地鼠卵中时,随着注入精子数量的增加,卵发育的异常率显著增加,而注射无顶体精子组,无论注射精子数量为多少,均无异常发生。②将6 ̄9个完整顶体的人精子注入人卵母细胞中时,未观察到发育异常的卵。此外,注射多个精子于金黄地鼠卵中时,多原核(PN)形成率顶体完整组明显低于无顶体组(P<0.01)。结论:本研究初步证实注射完整顶体人精子所含顶体酶对胚胎发育存在一定的危害性,行人类辅助生殖技术ICSI时,注射无顶体精子似乎更合理,或许能增加该技术的有效性。Objective: To investigate potential hazard of injecting acrosome-intact sperm to the embryo development when performing intracytoplasmic sperm injection(ICSI).Methods: 1) Injected 1,3,5 acrosome-intact sperm(s) into golden hamster oocytes,respectively.Observed oocyte activation,fertilization and post-fertilization development.Acrosome-removed sperm injection with equal numbers served as the control;2) Injected 6-9 acrosome-intact sperms into disposal human oocytes.Observed oocyte and embryo development.Results: 1) The rate of abnormal fertilized golden hamster oocytes significantly increased along with increasing number of acrosome-intact spermatozoa injected.No abnormal fertilized egg was found in the control whatever the number of sperm injected.Moreover,the incidence of polypronucleus formation was significantly lower in the acrosome-intact group than in the control(P0.01).2) No abnormality was found in human oocyte post-fertilization development with several acrosome-intact spermatozoa injected.Conclusion: Injection of acrosome-intact sperm possesses potential hazard to embryonic development,probably due to acrosomal emzymes contained in the acrosome.Injection of acrosome-removed spermatozoon appears more rational and effective when performing ICSI.
