摘 要:目的 观察去甲硫氨酸全肠外营养对大鼠血清氨基酸谱等的影响。方法 Wistar大鼠24只,随机分为含甲硫氨酸(+ MetTPN,n = 12)和去甲硫氨酸全肠外营养2 组(- MetTPN,n = 12),分别给予相应的TPN 支持。治疗7 d 后,每组随机抽取6 只大鼠处死,检测血清FAA(HPLC法)、肝肾功能和全血常规,同时作心、肺、肝、肾组织病理学检查。两组其余大鼠继续原TPN 治疗,观察生存期。结果 - MetTPN 组大鼠血清游离Met、Cys明显降低,Asp、Glu、Ser等显著增高;大鼠体重下降;血清总蛋白和白蛋白水平下降;血常规和肾功能未见明显异常;组织学检查见肝细胞轻度肿胀,细胞核仁增粗,心肺肾未见明显异常;平均生存18 d。对照组上述检查未见异常,除一只大鼠因导管并发症于TPN第16 d 死亡,其余大鼠全部存活。结论 - MetTPN 一周可致大鼠血清游离Met、Cys明显降低和Asp、Glu、Ser等显著增高,及轻度肝功能改变;随着- MetTPN时间延长。Objective To observe the effects of L methionine deprived total parenteral nutrition on the serum free amino acids of Wistar rats.Methods 24 male Wistar rats were divided into methionine contained (+MetTPN,n=12) and methionine deprived total parenteral nutrition (-MetTPN,n=12) groups radomly, and given TPN in different formula according to the group.After 7 days of the treatment, 6 rats in each group were sacrificed.The serum FAA, blood routine,liver and kidney functions,as well as the heart,lung,liver,kidney histopathlogical examinations were studied.The other animals in each group were continued to have their previous treatment until death.Results In the Met deprived group, plasma free Met and Cys levels decreased remarkably,and reversely,Asp,Glu and Ser levels increased significantly.The body weight,serum total protein and albumin also decreased.Blood routine and kidney function test were normal.The average life span was 18 days.There was no abnormal in above tests in the control group except one rat died of tube complication and the rest rats lived untill the experiment finished.Conclusion The serum free Met and Cys levels of the rats decreased remarkably after 1 week of methionine deprived total parenteral nutrition,however Asp,Glu and Ser levels increased to accompaniment of slight change of liver function, but as Methionine deprivation lasts enough time,all rats showed severe metablism disturbance and multiple organ failure and finally died.
