摘 要:目的从微量量热法角度出发,构建筛选中药抗菌活性部位的方法并探讨其可行性。方法以金银花为例,采用微量量热法,测定金银花不同部位对大肠杆菌的代谢影响,通过分析不同样品对大肠杆菌的热活性谱图(即热功率-时间曲线),得到一系列微量量热参数,分析金银花不同部位的药效作用及差异。结果金银花各部位对大肠杆菌的生长均具有抑制作用,且作用强弱存在显著性差异,通过主成分分析的方法,比较各部位的抗菌活性强弱。各部位抗菌作用强弱顺序为:总异绿原酸>总绿原酸>总黄酮>总环烯醚萜。结论微量量热法具有适用性广,简便快速的优点,该方法的运用为中药物质的药效初筛提供了一个新的研究思路。Objective To establish a novel method for screening bioactive parts and substances of traditional Chinese medicine based on microcalorimetry.Methods Using microcalorimetry,the power-time curves of Escherchia coli in different bioactive parts of folium of Lonicerae Japonicae were determined by TAM Air Isothermal Calorimeter.Quantitative thermokinetic parameters,such as k1,k2,t1m,t2m,P1m,P2m,Qsta,1,Qsta,2,and Qt,were obtained and the antibacterial effect of different bioactive parts was studied and compared.Results The different bioactive parts from Flos Lonicerae Japonicae had different antibacterial effects on Escherchia coli.Experimental results revealed that total isochlorogenic acid had the best antibacterial effect,followed by total chlorogenic acid,total flavone and total iridoid glycosides.Conclusion The microcalorimetric method has many virtues,such as extensive applicability and user-friendliness.
