摘  要:目的通过观察祛风活络丸对佐剂关节炎大鼠血清肿瘤坏死因子-α、白介素-1β水平的影响,探讨祛风活络丸治疗类风湿性关节炎的可能机制。方法以佐剂关节炎大鼠为实验模型,雷公藤多苷片为阳性对照药物,随机分6组(正常组、模型组、阳性对照药组、祛风活络丸低、中、高剂量组),采用放射免疫法检测大鼠血清中肿瘤坏死因子-α和白介素-1β的含量。结果佐剂性关节炎大鼠模型组血清肿瘤坏死因子-α、白介素-1β水平显著高于正常对照组;经祛风活络丸治疗后,祛风活络丸各剂量组大鼠血清肿瘤坏死因子-α、白介素-1β含量均明显低于模型对照组,其高、中剂量组均有非常显著性差异。结论祛风活络丸可能通过对细胞因子网络的调节,降低促炎性细胞因子肿瘤坏死因子-α和白介素-1β水平,抑制细胞因子的促炎效应,增加抗炎效应,从而减轻关节炎性细胞浸润及滑膜炎性增生。Objective To explore the possible role of Qufenghuoluo Pills in rheumatoid arthritis by detecting the expression of tumor necrosis facter-α,interleukin-1β in serum of adjuvant arthritis rats.Methods Adjuvant induced arthritis rat models were established and tripterygium glycosides used as positive control.Rats were divided randomly into normal group,model group,positive group,Qufenghuoluo pills-high group,Qufenghuoluo pills-middle group and qufenghuoluo pills-low group.The content of tumor necrosis facter-α,interleukin-1β in serum was determined respectively by RIA assay.Results Compared with the nomal group,the level of tumor necrosis facter-α,interleukin-1β in serum of model group was evidently increased.After drug treatment,the content of tumor necrosis facter-α and interleukin-1β Qufenghuoluo Pills of different-dosage groups was lower than that of model group,with that of high and middle dosage group dropped significantly.Conclusion Qufenghuoluo Pills can relieve inflammatory cells' infiltration and synovitis hyperplasia by regulating the cytokine network.It reduces inflammation promoting cytokine tumor necrosis facter-α,interleukin-1β and increased anti-inflammation cytokine to inhibit inflammation reaction.
