高效液相色谱法测定清心明目上清丸中盐酸小檗碱的含量

摘　　要：目的建立清心明目上清丸中盐酸小檗碱的含量测定方法。方法采用高效液相色谱法,色谱柱:AgilentZORBAX SB-C18(250 mm×4.6 mm,5μm),流动相:磷酸盐缓冲液〔0.05 mol/L磷酸二氢钾和0.05 mol/L庚烷磺酸钠(1∶1),含0.2%三乙胺,并用磷酸调节pH值至3.0〕-乙腈(70∶30);检测波长:345 nm;流速:1.0 ml/min;柱温:35℃。结果盐酸小檗碱在8.112～81.12μg/ml范围内呈良好的线性关系,平均回收率为96.59%,RSD为1.6%,n=5。结论本法可用于清心明目上清丸中盐酸小檗碱的含量测定,控制质量。Objective To determine the content of berberine hydrochloride in Qingxin Mingmu Shangqing Pills.Methods High performance liquid chromatography analysis was carried out using Agilent ZORBAX SB-C18（250 mm×4.6 mm,5 μm） and phosphate buffer-acetonitrile（73∶30） as the mobile phase.The buffer solution was a mixture of 0.05 mol/L solution heptanesulfonate（1∶1）,containing 0.2% of TEA.The phosphoric acid was adjusted to a pH of 3.0.The detection wavelength was 345 nm,the flow rate was 1.0 ml/min,and the column temperature was 35℃.Results The linear range was 8.112-81.12 μg/ml.The average recovery was 96.59% and RSD was 1.6%.Conclusion The method can be used for quality control of Qingxin Mingmu Shangqing Pills.
