摘 要:①目的建立测定大鼠血浆中五味子乙素浓度的HPLC方法。②方法采用乙腈沉淀血浆蛋白。HPLC的条件为:反相C18(4.6mm×150mm,5μm)色谱柱分离样品,以乙腈-水(78:22)为流动相,流速1.0mL/min,检测波长224nm,柱温23℃。③结果所建立方法在(0.50~50.0)μg/mL范围内线性关系良好(r=0.9975);平均回收率介于92.60%~103.90%,日内、日间相对标准偏差介于8.17%~12.50%。④结论所用方法简便,准确可靠,适用于五味子乙素大鼠血药浓度测定。Objective To establish a HPLC method for determination of γ-Scchisandrin in rats plasma.Methods Proteins in the plasma were precipitated by adding acetonitrile.The conditions of HPLC method determined:Separation of the samples was carried out on a reversed-phase C18 column(4.6mm×150mm,5μm) with a mobile phase of acetonitrile-H2O(78:22,v/v) at a flow rate of 1.0 mL.min-1.UV detection wavelength was set at 224nm and the temperature of column was kept at 23℃.Results The calibration curve was linear from 0.5~ 50.0μg.mL-1 with correlation coefficient of 0.9975.The recovery of assay was between 92.60% to 103.90%.Both intra-and inter-day coefficients of variation were between 8.17% to 12.50%.Conclusion A rapid and reliable method for determination of γ-Scchisandrin in rats plasma was developed.
