高效液相色谱法测定荷胆胶囊中牛磺熊去氧胆酸含量

摘　　要：目的建立测定荷胆胶囊中牛磺熊去氧胆酸含量的高效液相色谱法。方法采用Alltech Alltima C18色谱柱(250 mm×4.6 mm,5μm),流动相为用磷酸调pH至4.4的甲醇-0.03 mol/L磷酸二氢钠溶液(65∶35),流速1 mL/min,检测波长210 nm。结果牛磺熊去氧胆酸进样量在4.067 4～12.2μg范围内与峰面积线性关系良好(r=0.999 6),平均回收率为97.99%,RSD=1.43%(n=6)。结论高效液相色谱法快速、准确、有效,可用于荷胆胶囊中牛磺熊去氧胆酸的含量测定。Objective To establish an HPLC method for the assay of tauro-ursodesoxycholic acid in Hedan Capsule.Methods Alltech Alltima C18 column（250 mm×4.6 mm,5 μm） was adopted,the mobile phase was methanol-buffer containing monometallic sodium orthophosphate 0.03 mol/L（65 ∶35）（adjusted pH to 4.4 with ortho-phosphoric acid）,flow rate was 1 mL/min,detection wavelength was 210 nm.Results The linear range of tauro-ursodesoxycholic acid was with in 4.067 4-12.2 μg（r=0.999 6）.The average recovery rate was 97.99%,RSD=1.43%（n=6）.Conclusion The method is simple,rapid,accurate and can be usded in content determination of tauro-ursodesoxycholic acid in Hedan Capsule.
