糖原合成激酶3β表达与上皮性卵巢癌顺铂敏感性的关系研究

摘　　要：目的探讨肿瘤组织中糖原合成激酶3β表达与顺铂敏感性的关系以及降低SKOV3细胞中糖原合成激酶3β的表达逆转细胞顺铂耐药性的可能性。方法选取住院治疗的上皮性卵巢癌患者30例,所有患者采用TP方案进行化疗。采用免疫组化方法检测糖原合成激酶3β表达与临床疗效的关系。体外培养SKOV3细胞,采用氯化锂(LiCl)抑制糖原合成激酶3β,通过MTT法比较糖原合成激酶活性与细胞顺铂敏感性的关系。结果顺铂对不同糖原合成激酶3β表达活性的SKOV3细胞的IC50分别为(37.67±1.95)、(55.37±2.95)μmol.L-1,差异有显著性(P<0.05)。上皮性卵巢癌患者组织糖原合成激酶3β表达活性与TP方案的临床疗效相关,糖原合成激酶3β表达强阳性组有效率为71.4%,阳性组有效率为61.5%,阴性组有效率为30.0%。3组比较差异有统计学意义(P<0.05)。结论糖原合成激酶3β的表达与SKOV3细胞对顺铂的敏感性相关,不同糖原合成酶3β表达量与TP方案的临床疗效呈现相关性。Objective To explore the relationship between the expression of GSK-3 β and ovarian cancer sensitivity to cisplatin.Methods Thirty patients with epithelial ovarian cancer were selected and treated by Taxol and cisplatin.The expression of GSK-3 β was evaluated by immunohistochemistry.The relationship between the expression of GSK-3 β and the clinical efficacy was analysed.The SKOV3 cells were cultured and treated by LiCl.The cellular sensitivity to csiplatin was evaluated by MTT assay.Results The sensitivity to cisplatin of different cells was significantly different,with half inhibitory concentration of（37.67±1.95）and（55.37±2.95）μmol·L-1.The expression level of GSK-3 β was related to the clinical efficacy.The response rates of the strong positive expression group,the positive group and the negative group were 71.4%,61.5%,and 30.0% respectively.Conclusion The expression of GSK-3 β is related to the SKOV3 cell sensitivity to cisplatin and the clinical efficacy of Taxol and cisplatin.
