3种不同洗液洗涤血小板的比较及其保存的体外实验研究

摘　　要：目的:观察3种不同的洗液(生理盐水,ACD-A保养液和PASⅢM)对机采血小板质量和功能的影响;同时观察洗涤血小板保存于PASⅢM和M-sol后3 d其功能及活化程度的变化。方法:机采血小板分别用生理盐水,ACD-A保养液和PASⅢM洗涤血小板,洗涤后分别保存于PASⅢM和M-sol中观察0、24、48、72 h血小板的功能变化及激活程度的差异。用血小板计数仪测血小板数量,PMV、PDW;用比浊法测血小板聚集性;用流式细胞仪测血小板表面CD62P的表达情况。结果:①PASⅢM组CD62P的激活率较低,与其他2组分别相比差异有统计学意义(P<0.05)。②洗涤后血小板分别保存于PASⅢM和M-sol保养液中保存24 h和48 h后,所有的检测数据与对照组相(0 h)比较差异无统计学意义,2组之间比较差异无统计学意义,且在正常的范围之内;③保存72 h后血小板聚集性与对照组相比差异有统计学意义(P<0.01),2组之间比较差异无统计学意义;而血小板CD62P的激活率在2种保存液之间差异有统计学意义。结论:3组洗液比较,PASⅢM对血小板功能及激活程度影响最小,是较理想的洗液;在PASⅢM和M-sol保存液中保存48 h,血小板的质量指标仍在正常范围之内;洗涤血小板72 h,M-sol保存液中血小板的功能指标优于PASⅢM中的。Abstract Objective：To compare the effect of three kinds of solution（NS, ACD-A, PASⅢ M） , he quality and function of the plateletapheresis, and observe the changes of the washed platelets preserved in PASⅢ M and M-sol after three days. Method： Platelet concentrates（PCs） were washed by NS, ACD-A and PASⅢ M： and preserved in PASIII M and M sol. After 24 h, 48 h and 72 h,the platelets＇function and CD62P＇activation were tested respectively. The platelet count,MPV and MCHC were tested by Blood Cell Counter. CD62P was detected by flow cytometry. Result：① The CD62P activation rate in the group washed by PASⅢ M was less than that of the other groups,which had statistical significance（P〈0.05）. ②The washed PCs were preserved in PASⅢ M and M-sol for 24 h and 48 h,all data detected had no differences between two groups or control group （Oh）,meanwhile the count or function were in the normal range. ③ After the PCs preserved for 72 h,the piatelet coagulation was lower than that of the Oh group（P〈0.01）and the CD62P in the M-sol was lower than that in the PASⅢ M（P〈0.05）. Conclusion：Comparison among the solution groups showed that PASⅢ M was the best washing water which affected the function and the activated degree weaker than the others. The quality parameters of the PCs were in the normal field preserved in M-sol and PASⅢ M after 48 h. After the washed PCs were preserved for 72 h,the function of the platelet in M sol was better than that in the PASⅢM.
