摘 要:研究了培养基中添加乙酸钠、尿素以及Fe3+对三角褐指藻(Phaeodactylum tricornutum)生长和油脂积累的影响。结果表明,培养基中添加乙酸钠作为碳源的兼养生长模式对三角褐指藻的生长和油脂含量的增加有明显的促进作用(P<0.05),在乙酸钠浓度为5.0 g/L时得到最大生物量为1.28 g/L,是对照的1.7倍;而在乙酸钠浓度为0.5 g/L时油脂含量达到最大(28.90%),是对照的1.9倍;培养基中添加适当浓度的尿素作为氮源,可以显著地提高三角褐指藻的生长速率(P<0.05),但对生物量和油脂含量没有显著影响(P>0.05);培养基中Fe3+浓度的增加对三角褐指藻的生长速率、生物量及油脂积累均没有显著影响(P>0.05)。Effects to the growth and oil accumulation in Phaeodactylum tricornutum were investigated using mixotrophic culture independently added sodium acetate,urea and Fe3+.The results showed that there was significant increasing of growth and oil accumulation using the sodium acetate as a carbon source,and the maxmium biomass appeared as 1.28 g/L when used the culture with 5.0 g/L sodium acetate and showed 1.7 times to the control.The maximum oil content in P.tricornutum cells was 28.90% with 0.5 g/L sodium acetate,and which was 1.9 times to the control.The growth rate significantly increased when used the urea as the nitrogen source(P0.05),but the biomass and the oil content of the P.tricornutum were not obviously changed(P0.05).There was no effect to the growth rate,biomass and oil content of P.tricornutum by adding Fe3+ in culture.
