北京地区与手足口病相关的非EV71、非CoxA16型肠道病毒的分子特征分析  被引量:34

摘  要:目的了解北京地区引起手足口病的非EV71、非CoxA16型肠道病毒的病原构成及优势型别VP4区遗传特征分析。方法收集2009年4月.12月检测为非EV71、非CoxA16型肠道病毒的手足口病患者咽拭子标本共45份。提取病毒核酸,用巢式RT-PCR法扩增病毒VP4区序列,对PCR阳性扩增产物进行测序,通过与GenBank收录的序列BLAST比对确定病毒型别;并对优势型别进行序列和进化分析。结果45例标本中共有32例标本巢式RT—PCR检测为阳性(71%),其中26份PCR产物测序成功,经BLAST比对后,26例标本病毒型别均得到确定:CoxA10为11例;CoxA5为5例;CoxA12和CoxA4各2例;CoxA2、CoxA6、CoxA9、CoxA16、CoxB5、Echo11各I例。CoxA10在型别确定的非EVT1、非CoxA16型肠道病毒中占44%,为优势肠道病毒型别。11株CoxA10在VP4区核苷酸序列同源性为82%-99%,氨基酸序列同源性为96%.100%。进化分析显示,10株北京株CoxA10与中国广东株在同一进化树分支上,1株与CoxA10日本株在同一分支上。结论北京地区引起手足口病的肠道病毒型别多样,CoxA10为非EV71、非CoxA16型肠道病毒中的优势型别,北京地区CoxA10株包括两个不同的基因型。Objectives To explore the pathogenic spectrum of hand, foot and mouth disease in Beijing and to analyse the genetic characteristics of the VP4 region of the predominant type of non-EVT1, non- CoxA16 enteroviruses. Methods 45 HFMD specimens tested positive for enterovirus and negative for EV71 and CoxA16 were collected from April to December of 2009. The virus RNA was extracted, and the VP4 sequence was amplified by nested RT-PCR. The PCR positive products were sequenced and the enterovirus type was identified by sequence Blast. The sequence and phylogenetic analysis of the predominant virus type was performed. Results 32 out of 45 (71%) specimens were tested positive by nested RT-PCR, among which 26 PCR products were successfully sequenced. And the types of the 26 enterovirus strains were identi- fied by sequence Blast : 11CoxA10, 5 CoxA5, 2 CoxA12, 2CoxA4, 1CoxA2, 1CoxA6, 1CoxA9, 1CoxA16, 1CoxB5 and 1Echol 1. CoxA10 accounted for 44% in typed non-EVT1, non-CoxA16 enteroviruses, and was the predominant type. The 11 CoxA10 strains shared the nucleotide identities of 82%-99% and the amino acid identities of 96%-100% with each other in VP4 region. Phylogenetic analysis showed that 10 CoxA10 strains were clustered with Guangdong CoxA10 isolates and one clustered with Japan isolates. Conclusions Hand, foot and mouth disease in Beijing was concerned with diverse enteroviruses, and CoxA10 was the predominant type among non-EV71, non-CoxA16 enteroviruses; The CoxA10 strains isolated in Beijing could be clustered into two genotypes.
