复尔康注射液对人肝癌细胞株及裸鼠移植瘤的抑制作用

摘　　要：目的:研究复尔康注射液对人肝癌细胞株增殖及裸鼠移植瘤生长的影响。方法:采用CCK8试剂盒检测复尔康注射液对人肝癌细胞株SMMC-7721和BeL-7402增殖的抑制作用;建立SMMC-7721细胞裸鼠移植瘤模型,通过比较肿瘤体积(tumor volume,TV)、相对肿瘤体积(relative tumor volume,RTV)和相对肿瘤增殖率(T/C)考察复尔康注射液对肿瘤生长的影响。结果:复尔康注射液对SMMC-7721和BeL-7402的IC50分别为(61.51±1.58),(68.39±7.16)mg·L-1;不同剂量复尔康注射液能够不同程度地抑制裸鼠移植瘤的生长,与阴性对照组比较,P<0.05。低、中、高剂量复尔康注射液组(0.065,0.130,0.260 g·kg-1)T/C分别为57.75%,39.9%和44.08%。结论:复尔康注射液体外能抑制人肝癌细胞株的增殖,体内能明显抑制SMMC-7721细胞裸鼠移植瘤的生长。Objective： To investigate the effect of Fu-er-kang injection on the growth of hepatocellular carcinoma cells in vitro and tumor xenograft in vivo in nude mice.Methods： The cytotoxic effect of Fu-er-kang injection on hepatoma carcinoma SMMC-7721 and BeL-7402 cells was determined by CCK8 assay（cell counting kit-8）.After SMMC-7721 cells were transplanted into BALB/c-nude mice,inhibitory effect of Fu-er-kang injection on tumor growth was evaluated by measuring tumor volume（TV）,relative tumor volume（RTV） and relative tumor proliferation ratio（T/C）.Results： The IC50 values of Fu-er-kang injection on SMMC-7721and BeL-7402 cell lines were（61.51±1.58） and（68.39±7.16） mg·L-1,respectively.After treatment with Fu-er-kang injection（0.065,0.130 and 0.260 g·kg-1）,T/C were 57.75%,39.9% and 44.08%,respectively,in nude mice.Conclusion： Fu-er-kang injection can inhibit the proliferation of SMMC-7721 and BeL-7402 cells in vitro,and also effectively inhibit the growth of tumor xenograft（transplantation of SMMC-7721 cells） in vivo in nude mice.
