摘  要:目的:建立离体牛蛙肺泡膜模型并初步探讨胰岛素的肺吸收机制。方法:建立离体牛蛙肺泡膜模型,对模型的完整性、通透性及影响因素等进行考察,并初步研究胰岛素的肺吸收机制。结果:在4 h内,膜的紧密性和通透性良好,在一定范围内胰岛素透过牛蛙肺泡膜是非浓度、非方向依赖性的,且单体较六聚体透过率高。结论:离体牛蛙肺泡膜模型可作为评价胰岛素肺吸收的体外模型。经比较,单体胰岛素可能比六聚体胰岛素肺吸收好。Objective: To establish a Rana catesbeiana alveolar membrane model and to investigate the pulmonary absorption characteristics of insulin using this model.Methods: In the model of Rana catesbeiana alveolar membrane,the integrity,permeability and influence factors were investigated.The mechanism of insulin pulmonary absorption was studied.Results: During 4 h,the integrity and permeability of Rana catesbeiana pulmonary monolayer were satisfactory.Insulin permeated the alveolar membrane in a concentration-and direction-independent manner,and monomeric insulin was easier to be absorbed into the lung than hexameric insulin.Conclusion: Insulin can passively diffuse through the barrier of Rana catesbeiana alveolar membrane via a paracellular pathway.Rana catesbeiana alveolar membrane,as a transport model,has the potential to investigate the pulmonary absorption mechanism of insulin.
