海南地不容总碱的镇痛抗炎作用及毒性观察

摘　　要：目的研究海南地不容总生物碱的镇痛抗炎作用及急性毒性。方法采用小鼠热板法、扭体法及甲醛测痛试验观察镇痛作用;采用二甲苯致小鼠耳肿胀、醋酸致小鼠腹腔毛细血管通透性、棉球诱导小鼠肉芽组织增生试验模型观察抗炎作用,通过胸腔白细胞游走试验研究其对炎症渗出液中白细胞数量的影响,应用交叉菜胶致足肿胀试验测定炎性组织中前列腺素E(2PGE2)含量并初步探讨其抗炎机理,急性毒性试验采用改良寇氏法测定LD50。结果海南地不容总碱能明显延长小鼠舔后足时间,减少扭体次数,降低甲醛测痛试验第一、二时相的疼痛强度;减轻小鼠耳肿胀,抑制毛细血管通透性增高及肉芽组织增生,有效抑制白细胞游走,并能明显减少大鼠炎症组织中PGE2含量。其LD50为0.569g.kg-1,95%可信限为0.519～0.623g.kg-1。结论海南地不容总碱镇痛、抗炎作用显著,其机理可能与抑制PGE2的产生与释放有关。Aim To study the analgesic and anti-inflammatory effects of total alkaloids from Stephania hairtanensis H. S.Lo et Y.Tsoong in Hainan and it＇s acute toxicity in mice. Methods The analgesic effects were evaluated by the hotplate test,twisting reaction induced by acetic acid,and formalin test.The anti-inflammatory activity was tested by several typical inflammatory models involving dimethylbenzene-induced ear swelling in mice,capillary penetration method,cotton pellet granuloma. The anti-inflammatory mechanism was preliminarily investigated by examining the quantity of leukoeytes and level of prostaglandin E2 （PGE2）in the carrageenin-induced inflammatory diffusate. The LD50 and 95% creditability were calculated with developed Karber Method. Results The total alkaloids could effectively prolong the leg-licking time, decrease the twisting reaction, eounterwork the primary and secondary reaction of the pain intensity induced by formalin; It also could lighten ear edema,inhibit capillary permeability and the hyperplasia induced by cotton pellet,The quantity of leukocytes and PGE,. level in the carrageenin-induced inflammatory diffusate were considerably reduced. The LDs0 of it was 0.569g·kg-1 and the 95% creditability was 0.519-0.623g·kg-1 . Conclusion The total alkaloids have significant antiinflammatory and analgesia effects which may relevant to the inhibition of the production and release of the inflammatory factor PGE,.
