摘 要:目的:观察针刺对多发梗塞性痴呆大鼠海马NF-κB和IκB蛋白表达的影响,探讨针刺治疗多发梗塞性痴呆的作用机制。方法:通过栓子注入法制作多发梗塞性痴呆模型,造模成功大鼠随机分为模型组、针刺组和非穴组,并设正常组和假手术组。针刺处理3周后,运用免疫组化方法分析海马NF-κB和IκB的蛋白表达水平。结果:与正常组相比,模型组海马NF-κB及IκB蛋白表达显著升高。同时,针刺组大鼠海马NF-κB、IκB蛋白表达水平明显高于模型组。结论:针刺能够增强多发梗塞性痴呆大鼠海马NF-κB、IκB的蛋白表达水平,发挥神经保护作用,改善痴呆动物的学习记忆能力。Objective: To observe the influence of acupuncture on the protein expression of nuclear factor-κB(NF-κB) and inhibitor nuclear factor-kappa B(IκB) in hippocampus of multi-infarct dementia(MID) rats and explore the mechanism of acupuncture on MID.Method: The model of MID in rats was made by injecting homogeneous emboli into the internal carotid artery.The MID rats were randomly divided into model,acupuncture and sham-acupuncture groups.Meanwhile,normal and sham-operated rats were used as control.The protein expression levels of NF-κB and IκB in hippocampus of all rats were evaluated by immunohistochemistry method following a three-week period of acupunctural treatment.Result: A signi?cant increase in the protein expression of NF-κB was observed in hippocampus of rats in model group when compared with normal rats.Meanwhile,significant increases in the protein expression of NF-κB and IκB were found in hippocampus of rats in the acupuncture group when compared with the model rats.Conclusion: Acupuncture could exert neuroprotective effects and improve the abilities of learning and memory of MID rats by increasing the protein expression of NF-κB and IκB.
