黄精多糖干预长春新碱抑制骨髓基质细胞增殖的研究

摘　　要：目的:探讨黄精多糖(PSP)促进小鼠骨髓基质细胞(BMSCs)生长及干预长春新碱(VCR)对小鼠BMSCs增殖的抑制。方法:①PSP 0.5、1.0、2.0、4.0g/L,以及VCR 2.5、5.0、10、15mg/L分别与骨髓单个核细胞(BMMCs)共同培养14d,应用MTT法测定BMSCs增殖。②PSP 0.5、1.0、2.0、4.0g/L与BMMCs预培养2h后再分别加入终浓度为5.0mg/L的VCR继续培养14d,应用MTT法测定BMSCs增殖。结果:BMSCs培养14d后,PSP 1.0、2.0、4.0g/L组的OD值(3.80、4.48、4.05)均高于正常对照组(3.70),其中PSP 2.0g/L组OD值与正常对照组比较差异均有统计学意义(P<0.05)。VCR 5.0、10.0、15.0mg/L 3组OD值(2.36、1.83、1.67)均显著低于正常对照组(3.70)(P<0.05)。BMSCs培养第14d PSP 2.0g/L+VCR 5.0mg/L组OD值(3.51)与VCR 5.0mg/L组OD值(2.62)比较差异有统计学意义(P<0.05)。结论:VCR明显抑制BMSCs生长;PSP可促进BMSCs生长,且干预VCR对BMSCs增殖的抑制。Objective： To investigate the effect of polygonatum sibiricum polysaccharides（PSP） on the proliferation of bone marrow stromal cells（BMSCs） inhibited by Vincristine（VCR）.Methods：The bone marrow mononuclear cells（BMMCs） were cultured with PSP（2.5,1.0,2.0,4.0g/L） or VCR（2.5,5.0,10,15mg/L） in 37℃ 5%CO2 for 14 days,respectively and their effect on BMSCs proliferation was determined by MTT.The BMMCs were pretreated by PSP（2.5,1.0,2.0,4.0g/L） for 2h,then,cultured with VCR 5.0mg/L for 14 days,and the effect of PSP on BMSCs proliferation was determined by MTT.Results：The OD value increased during 1.0 g/L（3.80）,2.0 g/L（4.48） and 4.0g/L（4.05） groups of PSP in BMSCs culture for 14 days.There was statistical significance for OD value between 2.0g/L group（4.48） of PSP and control group（3.70）（P0.05）.The OD value decreased during 5.0mg/L（2.36）,10mg/L（1.83） and 15mg/L（1.67） groups of VCR in BMSCs culture for 14 days,and there was statistical significance between them and control group（3.70） for OD value（P0.05）.There was statistical significance between PSP 2.0g/L ＋ VCR 5mg/L group（3.51） and VCR 5mg/L group（2.62） for OD value（P0.05） after culturing 14 days.Conclusion： The results suggest that vincristine can inhibit the BMSCs proliferation.Polygonatum sibiricum polysaccharides promotes the BMSCs proliferation and prevents the growth suppression of BMSCs inhibited by vincristine.
