Structural Changes and Localization of Substance P and Neurofilament Immunoreactive Fibers in Intervertebral Disc of Ovariectomied Rats

摘  要:目的研究卵巢切除后鼠椎间盘结构及软骨P物质神经分布变化,探索骨质疏松性腰背部疼痛的原因。方法3月龄雌性SD大鼠64只,分组行双侧卵巢切除(OVX)及假手术(Sham-O);术后第1、3、5、7个月取L3-4、L4-5椎间盘。研究采用胶原VG特染,光、电镜观察及免疫组织化学PAP等方法。结果Sham-0组各阶段纤维环中胶原纤维结构排列规则,偶尔见到较小的串珠状SP阳性反应物。OVX组纤维环中胶原结构排列紊乱、断裂,以后部改变较明显,软骨细胞退变。椎间盘软骨及周围组织可见较多的SP免疫反应阳性神经末梢。结论OVX后椎间盘较早出现退行性改变,结构及功能的完整性破坏;SP的增加与局部炎症反应相关,可能与骨质疏松性腰背疼痛有关。Objective To study the correlations of structural changes and localization of substance P neurofilament fibers in ovariectomied rats and to understand the basis of osteoporotic back pain Methods Sixty four 3 months old female SD rats were divided into two groups, one underwent ovariectomy(OVX) and the other shamoperation (Sham-O) Sixteen animals in earch batch were sacrificed at 1, 3, 5 , 7th month to procure the L3-4, L4-5 intervertebral disc specimens for the examinations by means of collagen VG stain, microscopy, scanning, transmission electron microscopy and the immunohistochemical PAP of disc cartilage Results Great changes were seen in annulus fibrosus and nucleus of intervertebral disc such as breaking or rupture of collagen fiberlining, particularlly in the posterior aspect of the annulus fibrosus, chondrocytes showed degenerative changes as compared with Sham-O control group And the existence of SP immunoreactive nerve fibers in disc cartilage and annulus fibrosus from the first month, which existed as neurofilament with the orlentation following collagen fibers SP positive nerve fibers were present and increasing in quantity with aging through 7th month Also SP nerve fibers were present in disc surrounding tissue and related to the local inflammation Conclusions The integration of structure and function were broken and degeneration of intervertebral disc took place early after OVX; increase of SP was possibly related with local inflammatory process as well as with lower back pain in estrogen deficiency osteoporosis
