糖尿病大鼠肾脏转化生长因子β1与α3、β1整合素的相关分析

摘　　要：目的探讨糖尿病大鼠肾脏TGF-β1及α3、β1整合素的相关性及厄贝沙坦对其的影响。方法 30只8周龄雄性Wistar大鼠随机分为3组,分别为正常对照组(对照组,n=7)、糖尿病对照组(DM组,n=14)和厄贝沙坦干预组(DM+Irb组,n=9),一次性腹腔注射链脲菌素造糖尿病大鼠模型,造模成功4周后开始灌胃法给予相应计量厄贝沙坦干预8周。ELISA测定各组大鼠24尿白蛋白排泄率,PAS染色观察肾脏病理变化,免疫组化法半定量足细胞密度,实时荧光定量PCR检测各组大鼠肾脏TGF-β1、α3、β1整合素mRNA的表达。结果糖尿病大鼠肾脏TGF-β1mRNA的表达显著升高,伴α3、β1整合素mRNA的表达减少,足细胞密度降低,蛋白尿增加,厄贝沙坦干预后上述现象改善。结论糖尿病大鼠肾脏TGF-β1可能通过调控α3、β1整合素mRNA的表达,减少足细胞,抑制此途径可能是厄贝沙坦发挥肾保护作用的机制之一。Objective To explore the association between renal transforming growth factor-β1 （TGF-β1） and the expressions of α3 and β1 integrins and observe the effect of irbesartan on their expressions in diabetic rats. Methods Thirty 8-week-old male Wistar rats were randomly divided into normal control group （n=7）, diabetic control group （n=14） and irbesartan group （n=9）. Rat models of diabetes were established by a single peritoneal injection of streptozotocin （STZ）, and 4 weeks later the rats received irbesartan treatment for 8 weeks. Enzyme-linked immunosorbent assay （ELISA） was used to measure the urinary albumin excretion rate, and PAS staining was utilized to observe the renal pathologies. Immunohistochemistry was performed for semi-quantitative determination of podocyte density, and real-time RT-PCR was used to detect the renal TGF-β1 and α3/β1 integrin mRNA expressions. Results In diabetic rats, the expression of renal TGF-β1 mRNA was significantly increased, while α3 and β1 integrin mRNA expressions and podocyte density significantly decreased with increased proteinuria. Irbesartan obviously improved such changes. Conclusion In diabetic rats renal TGF-β1 can regulate α3 and β1 integrin mRNA expressions to reduce the number of podocytes, and inhibition of this pathway may be one of the mechanisms of the renal protective effect of irbesartan
