摘 要:目的观察螺内酯对快速心房起搏对家兔心房肌细胞L型钙离子通道亚基表达的影响。方法家兔18只随机分为3组,对照组、起搏组及螺内酯组各6只。起搏组和螺内酯组经颈内静脉将电极植入右心房,以600次/min行快速心房起搏8 h,起搏后应用半定量反转录-聚合酶链反应检测心房肌钙离子通道α1C亚基及β1亚基mRNA的表达。结果与对照组比较,起搏组钙离子通道α1C亚基及β1亚基mRNA表达明显减少,分别下降22%和26%,差异有统计学意义(P<0.01);螺内酯组钙离子通道α1C亚基及β1亚基mRNA表达降低明显减少,分别下降13%和17%,与起搏组相比,有统计学意义(P<0.05)。结论螺内酯可预防快速起搏后钙离子通道表达亚基的降低。Objective To investigate the effects of spironolactone on mRNA expression of the L-type calcium channel subunit of the atrium induced by rapid atrial pacing in rabbits.Methods 18 rabbits were randomly divided into 3 groups: control group(n=6),rapid pacing group(n=6)and spironolactone group(n=6),electrode catheters were positioned in the right atrium of rapid pacing group and spironolactone group.High-frequency atrial pacing of 600 bpm was started and maintained for 8 hours.Right atrium tissue was obtained for measurement of mRNA expression of L-typecalcium channel subunits by reverse transcription-polymerase chain reaction(RT-PCR).Results Compared with the control group,the mRNA of α1C and β1 of the rapid pacing group decreased 22% and 26%(P0.01);the mRNA of α1C and β1 of the spironolactone group only decreased 13% and 17%,compared with the rapid pacing group,there were significant differences(P0.05).Conclusion The spironolactone can attenuate the down-regulation of L-type calcium channel of atrium induced by the rapid atrial pacing.
