小檗碱、白头翁素对肠黏膜微血管内皮细胞E-selectin表达的影响

摘  要:为了观察内毒素即细菌脂多糖(lipopolysaccharide,LPS)对肠黏膜微血管内皮细胞(RIMECs)表面E-selectin表达的影响及小檗碱和白头翁素对E-selectin表达的调控作用,试验以体外培养的肠黏膜微血管内皮细胞为模型,采用免疫组化SP法检测不同浓度LPS(10μg/mL、5μg/mL、1μg/mL)、10μg/mL小檗碱、10μg/mL白头翁素刺激6 h后及10μg/mL小檗碱、10μg/mL白头翁素分别预孵1 h后加入1μg/mLLPS刺激6 h细胞表面E-selectin的表达情况。结果表明:E-selec-tin在正常RIMECs表面不表达,LPS刺激后E-selectin表达增加,不同浓度LPS对E-selectin表达无显著差异;白头翁素和小檗碱单独作用后内皮细胞E-selectin表达呈弱阳性,小檗碱和白头翁素预孵组E-selectin表达减少,经灰度分析与LPS组比较差异极显著。说明LPS可诱导E-selectin表达显著增加,小檗碱和白头翁素可下调LPS诱导的E-selectin表达增加,提示抑制内皮细胞E-se-lectin表达,从而抑制白细胞的贴壁和黏附是小檗碱和白头翁素抗炎机制之一。To evaluate the effect of lipopolysaccharide(LPS)on the expression of E-selectin in primary cultures of rat intestinal mucosa microvascular endothelial cells(RIMECs),and the regulation of berberine and anemonin on the expression of E-selectin induced by LPS.Focused on the effect of LPS(1 μg/mL,5 μg/mL and 10 μg/mL) on the expression of E-selectin from RIMECs,the effects of anemonin and berberine on RIMECs expression of E-selectin in response to LPS were investigated.After LPS(0,1 μg/mL)stimulation for 6 h with or without 10 μg/mL berberine,anemonin pretreatment for 1 h,expression of E-selectin from unstimulated and stimulated RIMECs were assessed by immunohistochemical evaluations.The results showed that the LPS enhanced the expression of E-selectin.Berberine and anemonin significantly inhibited LPS-induced expression of E-selectin.It suggests that berberine and anemonin may exert their beneficial therapeutic action in intestinal inflammation,at least in part,by inhibiting the expression of E-selectin,preventing LPS-induced microvascular endothelial cell damage.
