摘 要:目的评价子宫肌层平滑肌细胞(myometrial smooth muscle cell,MSMC)诱导血管生成能力与孕激素的相关性。方法由大鼠子宫分离培养MSMC用于:①体外研究:受到不同孕激素浓度(10-13、10-11、10-9和10-7mol/L)作用的MSMC内VEGF水平;受到孕激素刺激的MSMC内孕激素受体水平变化;②在体Matri-gel结节血管形成实验对比诱导血管生成能力:MSMC与热坏死的相应细胞比较;加入及未加入孕激素的MSMC比较。上述各组n=12。结果 10-11mol/L孕激素作用下MSMC的VEGF水平明显升高(P<0.05),有明显增多的孕激素受体。Matrigel内MSMC引起血管生长作用,尤其较大直径血管,该血管生长作用在孕激素(10-11mol/L)作用下水平升高(血管总数及小动脉均P<0.05)。热坏死细胞组内没有明显的血管生长。结论 MSMC具有强大的诱导血管生长能力,适当的孕激素对该能力有放大作用。Objective To evaluate ability of inducing angiogenesis by myometrial smooth muscle cell(MSMC) and effect of progesterone on it.Methods MSMC were isolated from the adult rat uterus and cultured for two studies: ① Intracellular VEGF levels were measured in MSMC cultured with progesterone(P:10-13,10-11,10-9 and 10-7 mol/L).Progesterone receptor(PR) level was compared with the progesterone stimulation.② Blood vessel density was evaluated 8 days after different cells were injected with matrigel into the subcutaneous tissue of adult rats.Different cells groups were normal or hot damaged MSMC,MSMC or MSMC with progesterone(n=12 each group) were compared.Results MSMC expressed VEGF with levels significantly(P0.05) up-regulated by progesterone at an optimal dose of 10-11mol/L.PR was up-regulated with progesterone.MSMC induced significantly more blood vessels than control group,especially large-diameter vessels.There was no angiogenesis with hot damaged MSMC.This angiogenic effect was enhanced by progesterone(10-11mol/L,P0.05 for total blood vessel and small artery).Conclusion MSMC induces angiogenesis effectively and is enhanced by an optimal dose of progesterone.
