摘 要:目的探讨黄芩苷的抗肝纤维化作用及可能机制。方法应用四氯化碳(carbon tetrachloride,CCl4)灌胃给药建立大鼠肝纤维化模型,应用血清学及形态学方法评价黄芩苷的作用,应用定量PCR技术检测转化生长因子β1(transforming growth factor-β1,TGFβ1)的mRNA表达。结果黄芩苷可降低肝纤维化大鼠的血清;谷丙转氨酶(alanine aminotransferase,ALT)、谷草转氨酶(aspartate aminotransferase,AST)、透明质酸(hyaluronic acid,HA)、层黏连蛋白(laminin,LN)Ⅲ型前胶原(precollagenⅢ,PCⅢ)和Ⅳ型胶原(collagenⅣ,CⅣ)水平;抑制肝纤维化的病理进展;减少肝纤维化时TGFβ1的mRNA表达。结论长期应用黄芩苷具有抗大鼠肝纤维化作用,其机制与抑制TGFβ1的表达有关。Objective To investigate the effect of baicalin on liver fibrosis and the possible mechanism.Methods Rat liver fibrosis was induced by using carbon tetrachloride(CCl4).The serum levels of alanine aminotransferase(ALT),aspartate aminotransferase(AST),hyaluronic acid(HA),laminin(LN),precollagenⅢ(PCⅢ) and collagenⅣ(CⅣ) were enzymatically assayed.The fibrotic situation was histologically evaluated and the mRNA expression of transforming growth factor-β1(TGFβ1) was determined by real-time PCR.Results Long-term application of baicalin significantly lowered the aberrantly upregulated ALT,AST,HA,LN,PCⅢ and CⅣ in the rats of liver fibrosis.The pathological progression of liver fibrosis was also alleviated by administration of baicalin.Moreover,the mRNA lever of TGFβ1 was significantly inhibited by baicalin.Conclusion Baicalin exerts significant anti-fibrotic action in liver fibrosis rats,which may be explained by the suppression of TGFβ1 expression.
