摘 要:目的:探究亚急性酒精中毒大鼠的学习记忆与海马p-CEEB表达的相关性。方法:将60只大鼠随机分为4组,实验组用0.16 g/ml、0.32 g/ml、0.48 g/ml三种浓度酒精(15 ml/kg体重)进行灌胃,对照组用蒸馏水灌胃。各组分别在灌胃14 d、21 d、28 d后进行行为学检测(Morris水迷宫实验)、血酒精浓度监测和病理学观察。结果:①酒精染毒14 d,高浓度组大鼠潜伏期增加(P<0.05);酒精染毒21 d、28 d时3个实验组大鼠潜伏期增加,中、高浓度组2 min内寻求正确次数减少、目标时间/总时间下降低(P<0.05);②酒精染毒14 d中、高浓度组和酒精染毒21、28 d 3个实验组大鼠海马内p-CREB阳性细胞数较对照组明显下降(P<0.05);③血液酒精浓度与潜伏期呈正相关,与寻求正确次数呈负相关,与阳性细胞数呈负相关,且各组间的差异具有统计学意义(P<0.05)(除酒精染毒14 d大鼠血液酒精浓度与寻求正确次数外)。结论:亚急性酒精中毒对学习记忆的损伤程度与染毒时间、血液酒精浓度呈直线正相关,可能通过降低海马p-CREB蛋白表达来影响大鼠学习记忆能力。Objective:To investigate the correlation between learning and memory and expression of p-CREB in the hippocampus of rats suffered from sub-acute alcoholism. Methods:Sixty rats were randomized into four groups. The model rats were developed by intragastric administration of alcohol at divided dose of 0.16 g/ml, 0.32 g/ml and 0.48 g/ml according to the protocol of 15 ml/kg, and control group were treated with intragastric administration of distilled water. The rats in each group were exanained after gavage by behavior test ( Morris water maze test), with detection of the blood alcohol concentration and observation of the pathological changes at day 14, 21 and 28, respectively. Results:(1)The latency was increased at day 14 in rats exposed to high alcohol (P 〈0.05) and in animals of the 3 trials at the 21st and 28th day. The emTect spatial start positions within two minutes were found declined in rats exposed to moderate and high level of alcohol besides reduction of target time / total time (P 〈 0.05). (2)p-CREB positive cells in hippoeampus were significantly reduced at day 14 in rats by moderate and high exposure to the alcohol and at the 21st and 28th day for aninmls of the three experimental groups ( P 〈 0.05 ). (3)Blood alcohol concentration was positively correlated with the latency, but negatively with the correct number of searching and p-CREB positive cells (P 〈 0.05) , with exception of the findings at the14th day regarding the correlation of blood alcohol concentration and the correct spatial start positions for rats exposed to the alcohol. Conclusion:The learning and memory impairment from sub-acute alcoholism is positively involved in exposure time and concentration of blood alcohol, suggesting that the possible mechanism may be associated with down-regulated level of p-CREB protein in hippocampus.
