摘  要:目的观察复元胶囊对兔膝骨关节炎软骨中胰岛素样生长因子-I(IGF-I)、胰岛素样生长因子结合蛋白3(IGFBP3)表达的影响。方法采用右后肢伸直石膏固定法造模,将44只雌雄各半新西兰大白兔随机分为正常组、模型组、盐酸氨基葡萄糖组、复元胶囊低、中、高剂量组。正常组4只,其余各组8只。各药物组每天灌胃1次,持续6 w。免疫组织化学法检测IGF-I、IGFBP3表达。结果模型组IGF-I、IGFBP3表达积分明显高于正常组(P<0.01);复元胶囊各剂量组与模型组比较,IGF-I表达积分不同程度升高(P<0.05),而IGFBP3表达积分不同程度降低(P<0.01),高剂量组显著。结论复元胶囊具有防治膝骨关节炎作用,其机制可能与调节IGF-I、IGFBP3表达水平有关。Objective To observe the influence of FuYuan capsule on IGF-Ⅰ and IGFBP3 in cartilage of rabbit with knee osteoarthritis.Methods 44 healthy New Zealand big rabbits composed of male and female in equal were randomly divided into normal,model,glucosamine hydrochloride and different dose FuYuan capsule groups,4 in normal group and 8 in other groups.The right hind limbs of all model rabbits were fixed by plaster bandage in the straight situation.Drug groups were respectively fed with glucosamine hydrochloride and different doses of FuYuan capsule one times daily for 6 weeks.The levels of IGF-Ⅰ and IGFBP3 in cartilage were detected through immunohistochemistry.The positive cells fraction and tinctorial strength were used by scoring method to measure the levels of IGF-I and IGFBP3.Results Total score of IGF-I and IGFBP3 expressions in model group were higher than those of normal group(P0.01).Compared with FuYuan capsule groups,especially high-dose and model groups,the total score of IGFBP3 level was decreased significantly(P0.01),while the total score of IGF-Ⅰ level was increased(P0.05).Conclusions FuYuan capsule can play an important role in prevention and treatment of osteoarthritis,which could be concerned to regulating the levels of IGF-I and IGFBP3 in cartilage.
