摘 要:采用辛酸-硫酸铵法和亲和层析法两种方法纯化兔抗苏丹红I IgG。纯化产物通过BCA法、琼脂双向扩散、SDS-PAGE凝胶电泳试验进行蛋白浓度、效价及纯度的验证。实验结果表明,利用辛酸硫酸铵纯化的产物蛋白质浓度高于亲和层析法,但经亲和层析纯化的产物纯度和效价均优于饱和硫酸铵。因此亲和层析法是一种快速高效的纯化方法,纯化产物可适用于免疫学检测方法的研究,为今后食品中苏丹红添加剂的快速检测奠定了良好的基础。To compare the effects of caprylic acid-ammonium sulfate(CA-AS) method and affinity chromatography method for purifying the rabbit anti-Sudan I IgG.The purified products were tested in the protein content,relative molecular weight,and immune titer by BCA method,agar gel precipitation(AGP) assay,sodium dodecyl sulfate polyacrylamide gel electrophoresis(SDS-PAGE).The results show that the protein content of purified antibody by CA-AS is higher than that by the affinity chromatography method,but the purity and titer with affinity chromatography method are both higher than that with AS method.Therefore,the affinity chromatography method is a rapid and efficient method for antibody purification.The purified products are suitable for the study of immunology test method,and establish a better groundwork for the rapid detection for Sudan I in food in the future.
