摘 要:目的:观察蒲公英萜醇和乙酰蒲公英萜醇对胃癌AGS细胞增殖和凋亡的影响。方法:采用四甲基固氮唑盐(3-(4,5-di methylthiazol-2-yl)-2,5-diphenyltetrazolium bromide,MTT)法测定蒲公英萜醇和乙酰蒲公英萜醇对AGS细胞生长的影响,流式细胞仪检测蒲公英萜醇和乙酰蒲公英萜醇对AGS细胞细胞周期的影响,Annexin V-异硫氰酸荧光/碘化丙啶双染法测定蒲公英萜醇和乙酰蒲公英萜醇对AGS细胞凋亡的影响。结果:蒲公英萜醇对AGS细胞的生长具有较好的抑制作用,该作用呈剂量和时间依赖性。进一步研究发现,蒲公英萜醇可以阻滞细胞周期于G2/M期,与对照溶剂相比,110μmol/L的蒲公英萜醇能显著增加处于G2/M期的细胞数(P<0.05);蒲公英萜醇也可以促进细胞的凋亡,可使细胞早期凋亡率明显增加,其中110μmol/L的蒲公英萜醇能将早期细胞凋亡率从4.45%升高到10.29%,是空白对照的2.31倍。乙酰蒲公英萜醇对AGS细胞生长的抑制作用弱于蒲公英萜醇,具有阻滞细胞周期于G2/M期的趋势,但差异无统计学意义(P>0.05),诱导凋亡的作用亦不明显。结论:蒲公英萜醇显著抑制AGS细胞的生长,该作用通过阻滞细胞周期于G2/M期和促进细胞凋亡实现。乙酰蒲公英萜醇的作用弱于蒲公英萜醇。Objective:To investigate the effects of taraxerol and taraxeryl acetate on cell cycle and apoptosis of human gastric epithelial cell line AGS cells.Methods:The inhibitory effects of taraxerol and taraxeryl acetate at different concentrations on AGS cell growth were measured by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide(MTT)method and the concentrations of taraxerol and taraxeryl acetate to be used in following experiments were decided.Then,cell cycle analysis was performed by FACScan flow cytometry after culture with taraxerol or taraxeryl acetate.Annexin V-fluorescein isothiocyanate/propidium iodide staining was used to measure cell apoptosis.Results:Taraxerol significantly inhibited AGS cell proliferation in a dose-and time-dependent manner.Taraxerol arrested the AGS cells at G2/M stage.110 μmol/L taraxerol elevated the population of AGS cells arrested in G2/M phase compared with solvent(P0.05).Taraxerol also promoted early cell apoptosis in AGS cells.110 μmol/L taraxerol increased the early cell apoptosis rate from 4.45% to 10.29%,which was 1.31 times higher than that of the untreated cells.However,taraxeryl acetate had a lower inhibitory effect than taraxerol,and it showed a tendency of G2/M arrest and apoptosis promotion but with no statistical significance(P0.05).Conclusion:Taraxerol has inhibitory effects on AGS cell growth through inducing G2/M arrest and promotion of cell apoptosis.Taraxeryl acetate has less effect on cell cycle arrest and apoptosis of AGS cells than taraxerol.
