刺松藻多糖抗凝血及抗血栓作用的研究

摘　　要：目的研究刺松藻多糖抗凝血作用及对血小板聚集的影响。方法采用热水浸提乙醇沉淀法制备刺松藻多糖。家兔心脏采血,取血浆检测活化部分凝血酶时间(APTT),凝血酶原时间(PT)和凝血酶时间(TT);采用红外比浊法测定刺松藻多糖对血小板聚集的影响。采用胶原蛋白-肾上腺素诱导小鼠血栓模型实验,检测刺松藻多糖抗血栓作用。结果刺松藻多糖能显著延长家兔血浆APTT、PT和TT;显著抑制由二磷酸腺苷(ADP)引起的家兔血小板聚集,对胶原蛋白-肾上腺素诱导的小鼠血栓性偏瘫及死亡率均有明显降低。结论刺松藻多糖具有抗凝血、抑制血小板聚集和抗血栓作用。Aim To investigate the inhibiting effects of polysaccharide of Codium fragile on blood coagulation and platelet aggregation.Methods Polysaccharides of Codium fragile were extracted by using hot water extraction and ethanol precipitation.The active partial thromboplastin time（APTT）,pronthrombin time（PT）and thromboplastin time（TT）of rabbits were tested by clotting model in vitro.Infrared turbidimetric method was used to estimate the effect of platelet aggregation.Mouse model of thrombosis by injecting collagen-adrenaline was applied to observe the antithrombotic effects.Results The polysaccharides can significantly prolonged APTT,PT and TT,prominently inhibited the platelet aggregation induced by adenosine diphosphate（ADP）.The hemiplegy and death rate was decreased in mouse by injecting collagen-adrenaline.Conclusion Polysaccharide has anticoagulant and antithrombotic effect.
