摘 要:目的观察不同浓度乌拉坦增强康斯特保护液(HTK)液对大鼠离体心脏保存效果,筛选最佳的乌拉坦浓度。方法 36只SD大鼠心脏随机分为6组,分别用含有不同浓度乌拉坦的HTK液(mmol/L:0、10、20、40、80和160)灌注停搏并于4℃静止保存8 h。采用langendorff离体心脏灌注系统和多导生理仪测定停搏前及保存后再灌注60 min的心功能,指标包括左室发展压、心率、左心室内压、左室舒张末压、收缩和舒张变化率,比较心功能各指标恢复率,同时检测保存液中肌钙蛋白I(cTnI)含量。结果 20 mmol/L乌拉坦增强HTK液保存大鼠离体心脏的心功能恢复率较其它浓度组显著升高,80 mmol/L组左室舒张末压显著高于其他组,其它各组之间没有显著性差异;加入乌拉坦各组cTnI释放量显著低于对照组,各浓度组之间没有显著性差异。结论 20 mmol/L乌拉坦增强HTK液对大鼠离体心脏具有最佳的保存效果。OBJECTIVE To investigate the preservative effects of histidine-tryptophan-ketoglutarate(HTK) solution enhanced with different concentrations of urethane on isolated rat heart,and to find the optimal concentration of urethane in these processes.METHODS Thirty-six hearts from SD rats were randomized into 6 groups and arrested with HTK solution enhanced with different concentrations of urethane(0 mmol/L,10 mmol/L,20 mmol/L,40 mmol/L,80 mmol/L and 160 mmol/L) and statically preserved in the same solution respectively for 8 h in 4℃.Langendorff perfusion model was used to measure the hemodynamic recovery of the hearts after preservation.Troponin I(cTnI) concentrations in the preservation solution were also examined.RESULTS All hearts resumed heart beating in 2 min reperfusion with KH solution in 37℃.The hemodynamic parameters of hearts preserved with HTK solution enriched with 20 mmol/L urethane recovered better than other solutions.The amount of cTnI released in the preservation solution decreased in all the solution enriched with urethane compared with original HTK solution.CONCLUSION Isolated rat heart is better preserved with HTK solution enriched with 20 mmol/L urethane.
