小鼠显微操作针的制作技术

摘　　要：目的探讨转基因小鼠制作中显微注射时所用移卵管、持卵针和注射针的最佳制作方案。方法通过酒精灯烧制和手拉以及通过P-97拉针仪设置不同的参数将薄壁硼硅玻璃管拉制成不同口径的移卵管、持卵针和注射针。结果按本方案制作的移卵针,持卵针和注射针用于小鼠受精卵的显微注射,实验过程顺利,效果非常好。结论该方法适用于小鼠受精卵显微注射的移卵管、持卵针和注射针制作。Objective To investigate suitable methods to make the holding pipettes,injection pipettes and pipettes for embryo handling during the production of transgenic mice. Methods Borosilicate glass tubes were burned by alcohol lamp burner and pulled by hand or in different parameters of P-97 puller to make the different inner and outer diameters of pipettes. Results Holding pipettes,injection pipettes and pipettes for embryo handling had good effect for microinjection with the increased survival rate of fertilized eggs. Conclusion The method is suitable to make pipettes for microinjection.
