摘  要:目的探讨褪黑素(MT)、5-羟色胺(5-HT)对致痫大鼠大脑皮质一氧化氮合酶(NOS)表达的影响。方法用免疫印迹技术和图像分析技术观察经MT预处理后致痫大鼠行为学的改变与脑内NOS表达的变化,用免疫组织化学方法检测大鼠海马内5-HT含量变化。结果戊四氮(PTZ)组大鼠大脑皮质内NOS含量明显高于对照组,PTZ+MT组大鼠大脑皮质内NOS含量明显低于对照组,差异均有统计学意义(P<0.05)。PTZ组、PTZ+MT+MT拮抗剂Luz组大脑皮质内5-HT含量与对照组比较均减少,差异有统计学意义(P<0.05)。结论 MT、5-HT具有明显的抑制大鼠大脑皮质NOS表达的作用和抗痫效应。Objective To investigate the effect of melatonin and serotonin on expression of nitric oxide synthase(NOS) in cerebral cortex of epileptic rats. Methods The model of epilepsy was induced by melatonin in rats and the expression of NOS in cerebral cortex was detected by western blot analysis.The level of 5-HT in hippocampus was demonstrated with immunohistochemistry.Results The expression of NOS in cerebral cortex of epileptic rats was increased in melatonin group,compared with control group,and there was significant difference(P〈0.05);Compared with melatonin group,glucocorticoids could significantly decrease the expression of NOS in cerebral cortex(P〈0.05) Conclusion Melatonin and serotonin have inhibitory effect on the expression of nitric oxide synthase in cerebral cortex of epileptic rats.
