摘 要:目的研究水囊压迫大鼠颅脑造成损伤后星形胶质细胞的反应。方法将改装后的5F漂浮导管球囊部位全部插入颅内硬膜外后,注射0.5 m l无菌生理盐水入球囊使其扩张并维持压力2 h。于伤后不同时间将大鼠处死,取脑组织进行HE染色和胶质纤维酸性蛋白(G lial fibrillary ac id ic prote in,GFAP)免疫组化染色。结果伤后存活1 h组,可见GFAP阳性细胞散在于损伤区周围,但突起较短,反应性星形胶质细胞主要局限于创缘周围。伤后存活24 h组,GFAP表达的范围扩大,在创缘及其伤区周围的阳性细胞增多,胞体增大,突起增长,且染色加深。24 h在海马组织中已有GFAP阳性细胞表达。伤后存活3 d、6 d和12 d组,阳性细胞的表达继续维持在高水平,且细胞形态各异,胞体变化较大,在创缘出现突起少而粗短的GFAP阳性细胞,而在损伤区周围、海马以及脑干的区域阳性细胞突起粗大不规则,突起进一步增长,胞浆突起染色较深。12 d时,GFAP阳性细胞增多达到高峰,阳性细胞着色深,呈深棕黄色。结论水囊压迫造成脑组织损伤后星形胶质细胞的标志性蛋白GFAP表达出现规律性变化,表明水囊压迫的损伤模型可以模拟颅脑血肿的损伤。Objective To investigate the response of astrocytes to craniocerehral injury caused by water-sac pressure on rat brain. Methods The water sac entirely inserted into encephalic putamen of rat was filled with sterile physiological saline and maintained its size and pressure for 2 hours. Rats were sacrificed at indicated times and their brain tissue were collected to carry out HE staining and immunohistochemical staining. Results In the 1 h injury group, Glial fibrillary acidic protein (GFAP) positive cells with short pustutes were found around the injury area and most of reactive astrocytes aggregated at wound margin.. In the 24 h injury group, the extent of GFAP expression increased and most of positive (:ells showed larger size of bodies, increscent pustutes and deep staining. Meanwhile, the total number of positive cells increased and aggregated at would margin and injury area. GFAP was expressed in hippocampus cells after 24 h injury. In the 3 d, 6 d, and 12 d injury groups, GFAP expression maintained at a high level and these positive cells displayed diverse celluar bodyand pustute shapes. When injured for 12 days, the expression of GFAP was highest and the mount of GFAP positive ceils reached a peak. Conclusion The water-bag pressure-caused brain injury model can mimic the pathogenesis of craniocerebral hematoma injury.
