摘  要:目的探讨晚期糖基化终末产物(AGEs)对成纤维细胞(Fb)形态及增殖功能的影响。方法体外培养人皮肤成纤维细胞,并加入不同浓度的AGEs干预处理,高倍镜下观察Fb的形态变化并采用MTT比色法检测细胞相对数量。结果体外培养的Fb加入AGEs干预后,胞体突起短且小,且细胞数量较对照组明显减少(p<0.01),在一定范围内呈剂量依赖性。结论 AGEs可显著影响Fb的形态及增殖功能,这可能与糖尿病足创面愈合延迟密切相关。Objective To observe the effect of advanced glycation end products(AGEs) on the morphological character and proliferation of fibroblast(Fb).Methods Fb was cultured in vitro,exposing to different concentrations of AGEs,and then observe the morphologic alteration at high magnification and examine the proliferation of Fb by MTT colorimetry.Results Compared with the controls,Fbs with AGEs had less and shorter lamellipodia.The quantity of Fbs with different concentration of AGEs was significantly decreased(p0.01) within tested dose range in a dose dependent pattern.Conclusion AGEs could alter the shape and inhibit the proliferation of Fbs obviously.It may partly account for the delayed healing of diabetic foot ulcers.
