摘  要:目的探讨姜黄素对心肺转流(CPB)术后大鼠肾脏的保护作用。方法 50只雄性SD大鼠随机均分为5组:假手术组(A组)、模型对照组(B组)、载体组(C组)、低剂量姜黄素组(D组)和高剂量姜黄素组(E组),分别在CPB开始前、结束时、术后2、4、122、4 h采血检测血清肌酐(SCr)、胱抑素C、肿瘤坏死因子α(TNF-α),术后24 h取肾脏进行组织学检查和TNF-α、环氧化酶2(COX-2)及核因子κB(NF-κB)检测。结果在术后2、4、12和24 h,D组和E组SCr、胱抑素C、TNF-α及组织中NF-κB和COX-2的含量均明显低于B组,但均高于A组(P<0.05)。组织学检查显示B组的肾小管上皮细胞肿胀、胞浆内空泡形成、间质出血等病理变化;但姜黄素组的变化较轻(P<0.05)。结论姜黄素可能通过抑制NF-κB及炎症因子的表达而显著降低CPB后肾脏炎症反应。Objective To investigate the renal protective effect of curcumin preconditioning on acute renal damage induced by cardiopulmonary bypass(CPB) in rats.Methods Fifty male SD rats were randomly divided into five groups of A(sham operated),B(model control),C(vehicle control),D(low-dose curcumin treated) and E(high-dose curcumin treated) with 10 rats each.Blood samples were collected before CPB,at the end of CPB,and at 2,4,12,24h after CPB for detection of serum creatinine(SCr),cystatin C and tumor necrosis factor-α(TNF-α).Kidney tissues were harvested at 24h after CPB for determining TNF-α,cyclooxygenase-2(COX-2) and nuclear factor-κB(NF-κB).Results The concentrations of SCr,cystatin C and TNF-α in blood at 2,4,12 and 24 h after CPB,and COX-2,NF-κB in renal tissues were lower in groups of D and E than those in group B,which were higher in groups of D and E than those in group A(P〈0.05).Histopathologic findings of group B confirmed that there were renal impairments characterizedby tubular dilatation,vacuole formation and stroma bleeding,but the impairments of group D and E were relatively small.Conclusion Curcumin can effectively reduce CPB-induced renal damage probably through inhibiting the activation of NF-κB and cytokines secretion.
