摘  要:利用ISSR技术对白花泡桐38个种源的遗传多样性进行分析。从100个引物中筛选出9个能扩增出清晰带型并具多态性的引物,共扩增出95个条带,其中88条具多态性,多态位点比率为92.63%;在物种水平上,种源的多态位点比率在32.63%(江西抚州)~56.84%(广西梧州和江西九江)之间,平均多态位点比率为47.16%,不同种源的有效等位基因数(Ne)、Nei’s基因多样性指数(H)、Shannon’s信息指数(I)的平均值分别为1.3910、0.2424、0.3765,均表明种源具有丰富的遗传多样性水平。种源遗传分化系数(GGst)为0.3539,种源问的遗传变异占总遗传变异的35.39%,表明种源间出现遗传分化,种源具有较高的遗传变异水平,且遗传变异主要来源于种源内的个体间。遗传一致度在0.39~0.82之间,就反映出白花泡桐种源的遗传基础较宽,存在较大的遗传变异性。其中贵州省凯里市和广西省柳州市种源的亲缘关系最近,遗传一致度为0.82,湖南省衡阳市与浙江省诸暨市、贵州省镇宁县种源的亲缘关系最远,遗传一致度为0.39。UPGMA法聚类分析将38个种源分为三组。研究还表明种源间的遗传距离与地理距离相关不显著。The genetic diversity of 38 provenances of Paulownia fortunei were analyzed by using inter-simple sequence repeats (ISSR). Total 95 DNA fragments were amplified using 9 primers with unamgiguous unique polymorphic bands screened from 100 ISSR primers, polymorphic loci of which were 88 (RppB =92. 63%); At the species level, the percentages of polymorphic loci (PPL) among different provenances were between 32. 63% (Fuzhou, Jiangxi) and 56.84% (Wu Zhou, Guangxi and Jiu Jiang, Jiangxi ) with the average of 47. 16%, the mean values of effective number of alleles (Ne), Nei's gene diversity index (H) and Shannon' s Information index (I) among provenances were 1. 391 0, 0. 242 4, 0. 376 5, respectively, and all of these data showed an abundant genetic diversity. The remarkable genetic differentiation occurred among provenances, which could be concluded from the coefficient of gene differentia- tion (GGst =0. 353 9), and the genetic variation between provenances accounted for 35. 39%, indicating that the genetic variation between provenances was higher than that of within provenances. Genetic Identity ranged from 0. 39 and 0. 82, reflecting the relatively wide genetic basis and genetic variation existed among provenances. Kaili, Guizhou and Liuzhou, Guangxi had closest relationship (0. 82), Hengyang, Hunan and Zhuji, Zhejiang, Hengyang, Hunan and Zhenning County, Guizhou had far distant relationship. 38 provenances of P. fortunei could be divided into 3 major groups obviously based on the UPGMA cluster analysis. The test also illustrated there was no significant correlation between genetic and geographic distances among provenances.
