摘 要:目的:研究虫草菌液(HS)对高糖作用下体外培养的大鼠腹膜间皮细胞(RPMCs)转化生长因子β1(TGF-β1)和纤维连接蛋白(FN)表达的影响。方法:将原代培养的第3代RPMCs随机分为6组:对照组、1.5%葡萄糖组、2.5%葡萄糖组、单纯虫草组(10mg/ml)、1.5%葡萄糖+10mg/ml虫草组、2.5%葡萄糖+10mg/ml虫草组。同步培养72h后,以RTPCR法检测各组细胞TGF-β1和FN mRNA表达情况;ELISA法检测细胞上清液中TGF-β1和FN的蛋白质水平。结果:高糖作用下RPMC的TGF-β1、FN mRNA和蛋白表达水平均明显高于对照组(P<0.05),且增高的程度与葡萄糖浓度呈依赖关系。与相应浓度的高糖组相比,经虫草菌液干预后RPMC的TGF-β1、FN mRNA和蛋白表达水平均显著降低(P<0.05)。结论:虫草菌液可下调高糖作用下RPMCs的TGF-β1和FN的表达,具有抗腹膜纤维化的作用。Objective:To investigate the effect of Hirsutella sinensis(HS) on the expression of transforming growth factor-β1(TGF-β1) and fibronectin(FN) in rat peritoneal mesothelial cells(RPMCs) cultured in high glucose concentration in vitro.Methods:The third generation RPMCs from primary culture were divided randomly into 6 groups:control group,1.5% glucose group,2.5% glucose group,HS(10 mg/ml) group,1.5% glucose+HS(10 mg/ml) group,2.5% glucose+HS(10 mg/ml) group.After 72 h,the mRNA and protein expression of TGF-β1 and FN were determined by RT-PCR and ELISA respectively.Results:The mRNA and protein expression of TGF-β1 and FN increased significantly in RPMCs cultured with high glucose,compared with control group(P0.05),and accompanying with the increased concentration of glucose.As compared with high glucose group(1.5% or 2.5% glucose),the mRNA and protein expression of TGF-β1 and FN decreased significantly in high glucose with Hirsutella sinensis (10 mg/ml) group(P0.05).Conclusion:Hirsutella sinensis reduces the expression of TGF-β1 and FN induced by high glucose in RPMCs,and has the inhibited effect on peritoneal fibrosis.
